新疆哈萨克族肥胖儿童菌群人源化动物模型的建立及评价

【摘要】:目的利用无菌小鼠建立新疆哈萨克族肥胖儿童菌群人源化(HFA)动物模型,研究哈萨克族肥胖儿童的肠道菌群在无菌小鼠体内的定植规律,为深入研究哈萨克族儿童肥胖的病理生理机制提供了一种新的选择方式。方法收集新疆哈萨克族肥胖儿童新鲜粪便样本,选取20只无菌级C57BL/6J小鼠(雌雄各半),于无菌小鼠3周龄时,开始灌胃接种哈萨克族肥胖儿童粪便菌悬液,接种1个月后收集HFA小鼠新鲜粪便样本,利用PCR-DGGE技术进行检测,对DGGE图谱进行相似性UPGMA聚类分析,评价哈萨克族肥胖儿童肠道菌群在HFA小鼠体内的定植规律。结果 HFA小鼠的菌群平均丰富度S为哈萨克族肥胖儿童的0.76倍;HFA小鼠的菌群平均均匀度E及多样性指数H均低于哈萨克族肥胖儿童,但差异无明显统计学意义;HFA小鼠与哈萨克族肥胖儿童肠道菌群的总相似度为64%,而HFA小鼠之间相似度为73%~94%;大部分雌性HFA小鼠与雄性HFA小鼠在聚类分析图上分离。结论 (1)本实验成功利用无菌C57BL/6J小鼠模拟哈萨克族肥胖儿童的特征性肠道菌群环境;(2)该HFA动物模型是探索哈萨克族儿童特征性肥胖与肠道菌群关系的一种新的选择方式。
[Abstract]:Objective to establish a humanized HFA-animal model of obese children of Kazak nationality in Xinjiang by using sterile mice and to study the colonization of intestinal flora of obese children of Kazak nationality in sterile mice. It provides a new way to study the pathophysiological mechanism of Kazak children obesity. Methods fresh fecal samples of obese Kazakh children in Xinjiang were collected, and 20 sterile C57BL/6J mice (half male and half female) were selected and inoculated with fecal bacteria suspension of obese Kazakh children at the age of 3 weeks. Fresh stool samples of HFA mice were collected one month after inoculation and detected by PCR-DGGE technique. The similarity UPGMA cluster analysis of DGGE profiles was performed to evaluate the colonization of intestinal flora of Kazakh obese children in HFA mice. Results the average abundance of bacteria in HFA mice was 0.76 times of that in obese Kazakh children. The average homogeneity E and diversity index of HFA mice were lower than those of obese Kazakh children. However, there was no significant difference between HFA-mice and Kazakh obese children in intestinal microflora. The total similarity between HFA-mice and Kazak obese children was 64, while that of HFA mice was 730.The majority of female HFA mice and male HFA mice were separated from male HFA mice by cluster analysis. Conclusion 1) C57BL/6J mice were successfully used to simulate the characteristic intestinal microflora environment of obese Kazakh children. The HFA animal model is a new way to explore the relationship between characteristic obesity and intestinal flora in Kazakh children.
