实验猕猴愤怒郁怒表情模式识别系统的构建及其药理应用

【摘要】:目的构建实验猕猴愤怒郁怒表情模式识别系统(以下简称“模式识别系统”),为猕猴模型表征评价提供客观手段;完成愤怒郁怒情绪反应及其诱发PMS肝气逆、肝气郁证猕猴模型的制备及评价,反证模式识别系统,为探索愤怒郁怒致病机理提供适宜的载体平台;应用猕猴愤怒郁怒表情模式识别系统对猕猴情绪变化予以量化,采集猕猴血样及脑中枢指标进行神经介质检测,通过数学建模手段构建宏微观指标非线性方程,,探查面部表情和PMS肝气逆、肝气郁两证发病及药物干预可能微观机制。 方法全国各实验猕猴饲养基地采集海量猕猴表情图片,动物行为学专家对表情进行分类评分,建立猕猴表情图片数据库,依据《雌性实验猕猴情绪评价量表》(以下简称“量表”)建立评价标准,采用机器学习方法构建模式识别系统;应用社会层级压力应激法诱导猕猴出现情绪反应,然后依据量表筛选出愤怒郁怒情绪反应猕猴模型,选取上述猕猴配合择时挤压造模使猕猴出现PMS肝气逆、肝气郁表现,应用中药对PMS肝气逆、郁证进行干预。随后采用模式识别系统和量表评分辅助判别其表情行为,验证模式识别系统的有效性;采集上述不同组别猕猴月经周期不同时期血清,分析血清和不同脑区NE、DA、5 HT和GABA含量,结合系统评分结果,将不同时期微观指标与其表情得分拟合非线性曲线,以期揭示情绪与体内指标间可能关系,为探讨PMS肝气逆、肝气郁两证发病及药物干预可能微观机制提供实验数据支撑。 结果1、模式识别系统可较好识别猕猴愤怒、郁怒表情,其得分与专家评分相关性为0.959。2、使用量表对猕猴造模前后进行评分,其愤怒因子与郁怒因子于造模前后有显著变化且符合预期规律。而各类微观指标的升降也均符合PMS猕猴模型造模前后应有的表现。使用模式识别系统对猕猴表情进行评分后发现:愤怒情绪反应与PMS肝气逆猕猴愤怒表情得分均较对照组显著上升(P0.01),PMS肝气逆组又显著多于愤怒情绪组(P0.05),PMS肝气郁猕猴郁怒表情得分显著多于对照组和郁怒情绪反应组(P0.01),中药干预可显著降低PMS肝气逆、郁证猕猴愤怒、郁怒表情得分(P0.01)。3、猕猴微观指标与同时期表情得分呈较明显非线性关系,其中各指标与表情得分的拟合方程分别为(X代表表情得分,Y代表微观指标含量ng/g):NE: Y=8.044+0.035X+0.003X~2 (1.692E 5)*X~3, R~2=0.921; DA:Y=28.371+0.095X+0.009X~2 (5.689E 5)*X~3, R~2=0.575;5 HT: Y=21.843 0.203X+0.005X~2 (2.244E 5)*X~3, R~2=0.716。 结论模式识别系统效标关联效度为0.959,可较好识别猕猴愤怒、郁怒表情,达到统计学要求;应用该模式识别系统进行实验过程中我们发现愤怒郁怒不良情绪反应与血清单胺类神经递质波动变化有关,而PMS肝气逆、郁证发病则与血清及中枢单胺类神经递质和GABA含量密切相关。中药干预可通过改变上述物质含量而发挥疗效。而应用模式识别系统及检测技术所采集宏微观指标的数据拟合结果则揭示不同组别、不同期属猕猴证候表征与其体内指标动态改变相因性,上述非线性方程构建无疑对认识不良情绪反应及其引发/诱发情志病证和中药干预的复杂机制提供了可资借鉴的思路及方法,为干预情志病证药物开发平台构建提供了方法学支撑。
[Abstract]:Objective To construct a model recognition system ( hereinafter referred to as " pattern recognition system " ) in an experimental rhesus monkey , which provides an objective way for the characterization and evaluation of the rhesus monkey model .
In order to explore the mechanism of anger - depression and depression , a suitable carrier platform is provided for the research on the preparation and evaluation of liver - qi depression , the preparation and evaluation of the model of liver - qi stagnation syndrome , and the evaluation of the syndrome pattern recognition system .
In this paper , the emotion changes of rhesus monkeys were quantified by the expression pattern recognition system , and the blood samples and the indexes of the brain were collected . The nonlinear equations of the macro - microscopic indexes were constructed by means of mathematical modeling , the facial expressions and PMS liver - qi adverse , the liver - qi stagnation syndrome and the possible micro - mechanism of drug intervention were explored .

Methods To collect the emoticons of a large amount of macaca mulatta from the breeding base of the experimental rhesus monkeys in the whole country , and the animal behavior experts classify the expression and establish the database of the emoticons of the rhesus monkey , and establish the evaluation standard according to the emotion evaluation scale of the female experiment ( hereinafter referred to as the " scale " ) , and construct the pattern recognition system by using the machine learning method ;
Using the social - level stress - stress method to induce the emotional response of Macaca mulatta , and then , according to the scale , we screened the model of the emotional response of the angry depression , and selected the above - mentioned rhesus monkey to make PMS liver - qi adverse and liver - qi stagnation . Then use the traditional Chinese medicine to intervene the PMS liver - qi and depression syndrome . Then the pattern recognition system and the scale score were used to help judge the expression behavior and verify the validity of the pattern recognition system .
