ESR技术在氧化应激自由基检测及抗氧化剂筛选中的应用研究

【摘要】:随着自由基医学和自由基生物学的发展,人们逐渐认识到自由基与人体的多种生理病理现象之间有着密切的关系。从自由基角度,探索自由基损伤的预防、诊断和治疗具有非常重要的意义。本论文一方面从检测自由基损伤的角度,利用电子自旋共振(ESR)技术考察了N,N-二甲基甲酰胺(DMF)诱导小鼠急性应激损伤模型中抗坏血酸及抗坏血酸自由基的变化,并以此作为自由基生物标记物,探索出一种检测DMF诱导氧化应激自由基损伤的方法,对DMF诱发职业病的诊断及病理研究具有一定的意义。另一方面从预防和治疗自由基损伤的角度,对外源性抗氧化剂进行了筛选。考察了11种常见中草药提取物对1,1-二苯基-2-苦基肼自由基、超氧阴离子自由基和羟基自由基的清除能力,并运用紫外可见分光光度法、化学发光法以及电子自旋共振检测方法,综合评价其体外抗氧化活性。结果显示绝大多数中草药均具有较好的抗氧化活性。其中,丁香、老鹤草、大黄清除自由基的能力最强。同时检测结果表明,同种中草药对不同自由基的清除能力存在一定差异,对同一自由基运用不同的检测方法,结果的一致性也有所差异。化学发光法与电子自旋共振技术相比,在检测抗氧化剂清除活性氧自由基时更具有一致性。
[Abstract]:With the development of free radical medicine and free radical biology, people gradually realize that there is a close relationship between free radical and various physiological and pathological phenomena of human body. From the angle of free radical, it is very important to explore the prevention, diagnosis and treatment of free radical injury. In this paper, on the one hand, the changes of ascorbic acid and ascorbic acid free radicals in acute stress injury model of mice induced by N-dimethylformamide (DMF) were investigated by electron spin resonance (ESR) technique from the angle of free radical damage detection. As a free radical biomarker, a method of detecting oxidative stress free radical damage induced by DMF was explored, which is of significance for the diagnosis and pathological study of occupational diseases induced by DMF. On the other hand, exogenous antioxidants were screened from the angle of preventing and treating free radical injury. The scavenging ability of 11 kinds of Chinese herbal medicine extracts on 1-diphenyl-2-picrylhydrazine radical, superoxide anion radical and hydroxyl radical was investigated. The methods of UV-Vis spectrophotometry, chemiluminescence and electron spin resonance were used. Comprehensive evaluation of its antioxidant activity in vitro. The results showed that most Chinese herbs had good antioxidant activity. Among them, clove, old crane grass, rhubarb scavenging free radical ability is strongest. At the same time, the results showed that there were some differences in the scavenging ability of the same Chinese herbal medicine to different free radicals, and the consistency of the results was also different when different detection methods were used for the same free radicals. Compared with electron spin resonance (ESR), chemiluminescence method is more consistent in the detection of antioxidant scavenging of reactive oxygen radicals (Ros).
