生长素受体TIR1、ABP1在人参中的基因克隆研究

【摘要】:生长素是植物生长过程中所必需的一种植物激素,在植物激素中是最早被发现的,存在于植物界的所有植物中,并在植物生长过程中起重要的协同作用。在植物生理活动过程中扮演重要角色。生长素主要产生于植物的分生组织——根尖和茎尖中,其他部位——茎和叶也可少量产生。它影响植物的生长进程,如细胞分裂和伸长,叶片及根、花、果实等形成,影响植物的向性运动。最近的二十年,以模式植物拟南芥为材料,科学家们主要从生长素的受体这个方面进行加进研究。生长素受体的特性和生长素的感应机制是在生长素发现的一个世纪以来被热切关注的课题。在1980年,生长素结合植物蛋白的定位,显示了受体成员中包含有生长素结合蛋白Auxin Binding Proteinl (ABP1),无疑影响了生长素的分子应答。2005年,由Mark Estelle和Ottoline Leyser两个研究小组在多年研究工作的基础上,发现TIR1在IAA应答基因的表达调控中起作用,能构成SCFTIR1复合体的F-box蛋白。TIR1已被国际学术界确定为生长素的受体。AXR1蛋白参与生长素信号传导的调节,在学术上研究及报道较少,属生长素受体研究的最新内容。 人参细胞生长素的受体基因至今未见研究文献报道,本实验主要根据模式植物拟南芥已知的生长素受体基因TIR1、ABP1的保守序列,设计人参生长素受体基因的引物,克隆人参生长素受体基因;后将重组的目的基因转移到到穿梭载体PINT-HIS3NB上,并在大肠杆菌DH10B上进行表达,得到阳性单克隆菌株后,提取质粒,进行测序比对,确定人参生长素受体TIR1、ABP1的基因序列。研究通过对生长素受体ABP1和TIR1在人参中的基因克隆分析,目的是阐明人参中生长素的受体基因序列以及ABP1和TIR1受体在人参中的序列分布,为进一步研究人参生长素的信号传递模式及对人参生长生理活动的影响机理奠定研究基础。
[Abstract]:Auxin is an essential plant hormone in plant growth. It is first found in plant hormones and exists in all plants in the plant kingdom and plays an important synergistic role in plant growth. Play an important role in plant physiological activities. Auxin is mainly produced in the meristem and stem tip of plant, and in other parts-stem and leaf. It affects the growth process of plants, such as cell division and elongation, leaf and root, flower, fruit and so on, and affects the directional movement of plants. In the last two decades, using model plant Arabidopsis thaliana as a material, scientists have been studying auxin receptors. The properties of auxin receptors and the sensing mechanism of auxin have been paid close attention to since the discovery of auxin in a century. In 1980, the localization of auxin binding plant proteins showed that the receptor member contained the auxin binding protein Auxin Binding Proteinl / ABP1, which undoubtedly affected the molecular response of auxin. Based on years of research work by Mark Estelle and Ottoline Leyser, we found that TIR1 plays an important role in the regulation of IAA response gene expression. F-box protein. TIR1, which can form SCFTIR1 complex, has been identified as auxin receptor. AXR1 protein is involved in the regulation of auxin signal transduction. The receptor gene of Ginseng cell auxin has not been reported so far. The primer of the receptor gene of Ginseng auxin receptor was designed based on the conserved sequence of the known auxin receptor gene TIR1 / ABP1 in Arabidopsis thaliana. The recombinant target gene was transferred to the shuttle vector PINT-HIS3NB and expressed on the Escherichia coli DH10B. After obtaining the positive monoclonal strain, the plasmid was extracted and sequenced. The sequence of ABP1 gene of Ginseng auxin receptor TIR1 was identified. The aim of this study was to elucidate the sequence of auxin receptor genes and the distribution of ABP1 and TIR1 receptors in ginseng by cloning and analyzing the auxin receptor ABP1 and TIR1 in ginseng. The results provide a basis for further study on the signal transduction mode of Ginseng auxin and the mechanism of its effect on the growth and physiological activities of Panax ginseng.
