Notch信号在小鼠胚胎早期造血的表达

【摘要】:目的 探讨小鼠胚胎发育过程早期造血相关组织中Notch1和Notch4信号的基因与蛋白的表达情况。 方法 采用PT-PCR和Western-blotting方法,对小鼠胚胎9.5天(E9.5)、E10.5、E11.5、E12.5胚胎的主动脉-性腺-中肾(aorta-gonad-mesonephros , AGM)和胎肝Notch1和Notch4信号基因和蛋白的表达进行检测。 结果 Notch1 mRNA在小鼠胚胎AGM区表达随孕期延长逐渐降低。Notch1 mRNA在胎肝E10.5表达最低,E11.5表达最高。Notch1蛋白与Notch1mRNA表达规律相似。Notch1 mRNA在AGM与胎肝各时间点比较,均具有显著差异;Notch1蛋白表达除E9.5AGM区明显高于胎肝外,余时间点表达无明显差异。 AGM区Notch4 mRNA在E10.5表达最高;在胎肝中表达随孕期延长而降低。Notch4蛋白与Notch4 mRNA表达规律相似。Notch4 mRNA在AGM区与胎肝各时间点比较,在E9.5和E12.5有显著差异,在E10.5与E11.5无明显差异。 结论 Notch1和Notch4信号在小鼠胚胎早期造血组织中有表达。随着胚胎发育,AGM区的造血能力下降,Notch1信号随之减弱;相反,胎肝造血功能逐渐完善,Notch1信号逐渐增强,提示Notch1信号可能与小鼠胚胎早期造血发育相关。AGM区Notch4信号在E10.5表达最高,其后逐渐下降;胎肝Notch4信号随孕期延长逐渐降低,提示Notch4信号可能与胚胎造血组织相关。
[Abstract]:Purpose To investigate the gene and protein expression of Notch1 and Notch4 signals in hematopoietic related tissues during early embryonic development in mice. Method The expression of Notch1 and Notch4 signal genes and proteins in aorta gonad-mesonephros (AGMs) and fetal liver were detected by PT-PCR and Western-blotting. Result The expression of Notch1 mRNA in the AGM region of mouse embryo decreased gradually with the prolongation of pregnancy. The lowest expression of Notch1 mRNA in fetal liver E10.5 the highest expression of Notch1 protein was similar to that of Notch1mRNA. The expression of Notch1 mRNA in AGM was compared with that in fetal liver at different time points. There were significant differences in the expression of Notch1 protein except in the E9.5AGM region, but there was no significant difference in the expression of Notch1 protein at other time points. The expression of Notch4 mRNA in AGM region was the highest in E10.5, and the expression of Notch4 protein in fetal liver was similar to that in Notch4 mRNA. The expression of Notch4 mRNA in AGM region was significantly different from that in fetal liver at each time point, but there was no significant difference between E9.5 and E11.5 in E10.5 and E11.5. Conclusion Notch1 and Notch4 signals were expressed in early embryonic hematopoietic tissues of mice. With the decline of hematopoiesis ability of AGM region in embryonic development, the signal of Notch1 decreased, on the contrary, the signal of fetal liver hematopoietic function was gradually enhanced, suggesting that Notch1 signal may be related to the early embryonic hematopoietic development of mice. The Notch4 signal of AGM region is the highest in E10.5. Notch4 signal of fetal liver decreased gradually with the prolongation of pregnancy, suggesting that Notch4 signal may be related to embryonic hematopoietic tissue.
