肠系膜上静脉血栓形成大鼠模型的建立

【摘要】:目的运用结扎法建立肠系膜上静脉血栓形成大鼠模型,模拟该疾病的病理过程,为研究其发病机制和治疗方法提供基础准备。方法 96只雄性SD大鼠,随机分为三组,A组(假手术组)、B组(绞窄型组)和C组(单纯型组),每组大鼠32只。A组大鼠仅打开腹腔,不阻断血运,术后8、24、48、72 h分批处死大鼠;B组和C组大鼠分别运用结扎法建立肠系膜上静脉血栓形成绞窄型和单纯型模型,建模后8、24、48、72 h分批处死大鼠。HE染色观察大鼠肠道组织形态学变化并对损伤进行病理评分;ELISA法检测大鼠血清中肠脂肪酸结合蛋白(IFABP)和α-谷胱甘肽S转移酶(α-GST)水平。结果 HE染色和病理评分结果示,与A组大鼠相比,B组和C组大鼠肠道组织均出现不同程度的血液瘀滞和损伤,B组逐渐加重,C组逐渐减轻,且血液瘀滞和损伤程度与结扎范围呈正相关。血清ELISA法结果示,与A组大鼠相比,B组和C组大鼠血清中IFABP和α-GST水平均呈现不同程度的升高(P0.05),且升高程度与结扎范围呈正相关。结论本研究运用结扎法成功建立了肠系膜上静脉血栓形成大鼠模型,简单易行,且手术成功率高,可在相关研究中使用。
[Abstract]:Objective to establish a rat model of superior mesenteric vein thrombosis by ligation, and to simulate the pathological process of the disease. Methods 96 male Sprague-Dawley rats were randomly divided into three groups: group A (sham-operated group) and group C (group C). Rats in group B and group C were killed in groups B and C respectively to establish strangulation and simple mesenteric venous thrombosis models by ligation at 824 hours and 48 hours after operation. The rats were sacrificed in groups for 72 h after modeling. The pathological changes of intestinal tissue were observed by HE staining and the levels of IFABP and 伪 -GSTs in serum of rats were detected by Elisa. Results the results of HE staining and pathological score showed that the intestinal tissue of rats in group B and group C had different degrees of blood stasis and injury. Blood stasis and degree of injury were positively correlated with the range of ligation. The results of serum ELISA method showed that the levels of IFABP and 伪 -GST in serum of group B and group C were increased in varying degrees compared with group A, and there was a positive correlation between the level of elevation and the range of ligation. Conclusion the rat model of superior mesenteric vein thrombosis was successfully established by ligation.
