人甲型H7N9禽流感全病毒灭活疫苗在小鼠中的免疫原性

【摘要】:目的评价人甲型H7N9禽流感全病毒灭活疫苗在小鼠中的免疫原性。方法分别将H7N9与H1N1、H5N1全病毒灭活疫苗稀释至血凝素抗原(hemagglutinin,HA)浓度分别为30、6、1.2、0.24和0.048μg/ml后,免疫BALB/c小鼠,各10只,0.5 ml/只,采用血凝抑制(hemagglutinin inhibition,HI)试验检测免疫后小鼠血清抗体滴度,计算抗体几何平均滴度(GMT)及半数有效剂量(ED_(50)),并检测免疫后小鼠血清对其他亚型病毒的HI滴度。结果 H7N9组小鼠阳转率与H1N1组较相近,仅H7N9 0.6μg剂量组GMT明显低于H1N1同剂量组(P0.001),其余剂量组差异无统计学意义(P0.05);H7N9与H1N1、H5N1全病毒灭活疫苗免疫小鼠ED_(50)分别为0.27、0.12和15μg HA。免疫后血清对其他亚型病毒无交叉反应。结论 H7N9疫苗在小鼠中的免疫原性略低于H1N1疫苗,但明显优于H5N1疫苗。
[Abstract]:Objective to evaluate the immunogenicity of inactivated human avian influenza A (A H 7 N 9) vaccine in mice. Methods the hemagglutininine (HA) antigen-hemagglutininine (HA) concentration of H7N9 and H _ 1N _ 1 inactivated H5N1 vaccine were diluted to 30 ~ 61.2g ~ 0.24 渭 g/ml and 0.048 渭 g/ml, respectively. BALB / c mice were immunized with hemagglutinin inhibition (HI) test, and the antibody titer was detected by hemagglutinin inhibition test. The geometric mean titer of antibodies (GMTT) and the half effective dose of EDTs were calculated and the HI titers of serum against other subtypes of viruses were detected. Results the positive conversion rate of H7N9 group was similar to that of H1N1 group, but the GMT of H7N9 0.6 渭 g group was significantly lower than that of H1N1 same dose group (P0.001). There was no significant difference in other dose groups (0.270.12 渭 g vs 15 渭 g HA). After immunization, the serum did not cross react to other subtypes of virus. Conclusion the immunogenicity of H _ 7N _ 9 vaccine in mice was slightly lower than that of H1N1 vaccine, but significantly superior to that of H5N1 vaccine.
