温肾补虚方对甲减模型大鼠血脂及TSHR表达的影响

【摘要】：目的本实验通过温肾补虚方对甲减大鼠的血脂及TSHR表达的影响,探讨中医药治疗甲减引起血脂异常的机制及方法。方法雄性SD大鼠随机分为空白组、造模组,造模组予0.05%丙硫氧嘧啶(PTU)蒸馏水。造模成功后,造模组随机分为模型组、西药组、中低剂量组、中药中剂量组、中药膏剂量组,予相应药物,给药时间4周。免疫组化法检测TSHR的表达;ELISA法检测CHOL、LDL、TSH、T3、T4、FT3、FT4。结果造模后,大鼠血清CHOL、LDL、TSH、TSHR升高(P0.01),T3、T4、FT3、FT4降低(P0.01)。治疗后,与模型组相比:中药组、西药组T3、T4、FT3、FT4升高显著(P0.01);中药组CHOL、LDL显著下降(P0.05);中药组及西药组TSH极显著下降(P0.01);中药组TSHR水平显著下降(P0.01)。结论温肾补虚方能改善甲减大鼠CHOL、LDL、TSHR的含量。中药调节CHOL、LDL的机制可能与降低TSHR的表达水平相关。
[Abstract]:Objective to explore the mechanism and methods of treating hypothyroidism with traditional Chinese medicine (TCM) through the effect of Wenshen Bushu recipe on blood lipid and TSHR expression in hypothyroidism rats. Methods male Sprague-Dawley rats were randomly divided into blank group. The model group was treated with 0.05% propylthiouracil (PTU) distilled water. After the establishment of the model, the model group was randomly divided into three groups: model group, western medicine group, middle and low dose group, middle dose group and ointment group, and the corresponding drugs were given for 4 weeks. The expression of TSHR was detected by immunohistochemical method. Results after the establishment of the model, the serum level of TSHHR of CHOLL LDL was increased, and the level of T3, T4, FT3, FT4 and P0.01 was decreased. After treatment, compared with the model group, the T3T4FT3FT4 in the western medicine group increased significantly (P 0.01); in the traditional Chinese medicine group, the Choll LDL decreased significantly (P 0.05); the TSH in the Chinese medicine group and the western medicine group decreased significantly (P 0.01); and the TSHR level in the Chinese medicine group decreased significantly (P 0.01). Conclusion Wenshen Bu deficiency prescription can improve the content of LDL TSHR in hypothyroidism rats. The mechanism of Chinese medicine regulating CHOLLL-LDL may be related to the decrease of TSHR expression level.
