DhHP-6对线虫在热环境下的保护作用

【摘要】:次血红素六肽(Deuterohaemin-Ala-His-Thr-Val-Glu-Lys,DhHP-6)是本实验室设计并合成的一个过氧化物酶(HPX)的新型模拟酶。前期实验结果表明DhHP-6给药能够明显提高野生型秀丽线虫的平均寿命。 本实验用经典模式生物秀丽隐杆线虫为模型,进一步证明了DhHP-6能分别提高秀丽隐杆线虫在30°C与35°C热环境下的生存率与寿命,并且DhHP-6能显著提高30°C热激后短期内的HSP-16.1表达量,并根据基因表达量的变化提出了一个可能的DhHP-6对线虫热保护的机制。为进一步的研究过氧化物酶与抵抗热激之间的关系打下基础。
[Abstract]:Deuterohaemin-Ala-His-Thr-Val-Glu-Lys-LysDhHP-6) is a new mimic enzyme of peroxidase (HPX) designed and synthesized in our laboratory. The experimental results showed that DhHP-6 administration could significantly improve the average life span of wild nematode. In this experiment, we used the classical model biological nematode, DhHP-6, to improve the survival rate and lifespan of Rhizoma elegans at 30 掳C and 35 掳C, respectively, and DhHP-6 could significantly increase the expression of HSP-16.1 in the short term after 30 掳C heat shock, and DhHP-6 could increase the expression of HSP-16.1 in a short period of time after 30 掳C heat shock. A possible thermal protection mechanism of DhHP-6 against nematode was proposed according to the change of gene expression. It provides a basis for further study on the relationship between peroxidase and heat shock resistance.
