丹参酮对左氧氟沙星在盆腔炎模型大鼠子宫组织中分布的影响

【摘要】:目的:探讨丹参酮对左氧氟沙星在盆腔炎模型大鼠子宫组织中分布的影响。方法:选取96只盆腔炎模型大鼠,随机分为实验组和对照组,每组48只,实验组肌注丹参酮(8 mg/kg),对照组肌注相同体积生理盐水;1 h后,2组动物均按80 mg/kg灌服左氧氟沙星(Levofloxacin,LVFX),于灌服LVFX后的0.25、0.5、1.0、1.5、2.0、4.0、8.0、12.0 h不同时间点,2组动物分别处死大鼠6只,采集血浆和子宫组织。高效液相色谱法法测定血浆和子宫组织中LVFX浓度,DAS2计算药动学参数。结果:实验组和对照组血浆与子宫组织血药浓度-时时间曲线均为二室模型,主要药动学参数列举如下:血浆中峰浓度Cmax分别为(14.341±1.551)μg/m L和(12.780±1.062)μg/m L,子宫组织中C_(max)分别为(13.910±1.704)μg/m L和(11.671±1.134)μg/m L(P0.05);血浆中达峰时间tmax分别为(1.329±0.269)h和(1.249±0.264)h,子宫组织中t_(max)分别为(1.419±0.380)h,(1.280±0.270)h;血浆中药物的半衰期t1/2β分别为(6.284±1.132)h和(5.442±2.590)h,子宫组织中t1/2β分别为(7.920±2.802)h和(3.894±1.595)h(P0.05);血浆中曲线下面积AUC0→t分别为(62.667±4.516)μg·h/m L和(55.693±3.138)μg·h/m L(P0.05),子宫组织中AUC0→t分别为(55.518±11.709)μg·h/m L和(40.374±2.892)μg·h/m L(P0.05)。结论:丹参酮能明显增加盆腔炎模型大鼠子宫组织中左氧氟沙星的分布。
[Abstract]:Aim: to investigate the effect of tanshinone on the distribution of levofloxacin in uterus of rats with pelvic inflammatory disease. Methods: 96 rats with pelvic inflammatory disease were randomly divided into experimental group and control group with 48 rats in each group. The experimental group was injected with tanshinone (8 mg/kg), the control group was injected with the same volume of normal saline for 1 h and the two groups were given Levofloxacin (LVFX) at 80 mg/kg. Plasma and uterine tissue were collected. Determination of LVFX concentration in plasma and uterine tissue by high performance liquid chromatography (HPLC) DAS2 was used to calculate the pharmacokinetic parameters. Results: the plasma concentration-time curves of plasma and uterine tissue in the experimental group and the control group were all two-compartment models. The main pharmacokinetic parameters were listed as follows: plasma peak concentration C max was (14.341 卤1.551) 渭 g / mL and (12.780 卤1.062) 渭 g / mL, C _ (max) in uterine tissue was (13.910 卤1.704) 渭 g / mL and (11.671 卤1.134) 渭 g / m / L (P0.05), plasma peak time (tmax) was (1.329 卤0.269) h and (1.249 卤0.264) h, t _ (max) in uterine tissue was (1.419 卤0.380) h, (1.280 卤0.270) h, in plasma drug concentration was (1.329 卤0.269) h and (1.249 卤0.264) h, respectively. 鐨勫崐琛版湡t1/2尾鍒嗗埆涓,
