脂肪移植治疗DNCB诱导ACD模型小鼠的实验研究

【摘要】:目的探究脂肪移植治疗2,4-二硝基氯苯(DNCB)诱导变应性接触性皮炎(allergic contact dermatitis,ACD)模型小鼠的效果及其可能机制。方法采用DNCB复制ACD小鼠模型,随机分成A(脂肪移植治疗组)、B(假手术治疗组)、C(药物治疗组)、D组(自然修复组),观察脂肪移植对ACD小鼠的治疗效果。2周后处死小鼠,取背部皮损处全层皮肤组织,苏木精-伊红染色法染色观察各组小鼠皮肤组织病理改变;实时荧光定量聚合酶链反应(q RT-PCR)检测皮损中白介素10(IL-10)、白介素17(IL-17)、干扰素-γ(IFN-γ)的表达。结果 A、C组可以观察到表皮厚度恢复正常,角质层变薄;B、D组无明显改变。RT-PCR检测发现A、C组皮肤组织中IL-10和IL-17 m RNA水平低于D组,IFN-γm RNA较D组升高(P0.05),而B与D组比较差异无统计学意义(P0.05)。结论脂肪移植可降低皮损中IL-10和IL-17的表达,促进IFN-γ产生,减轻皮损炎症。
[Abstract]:Objective to investigate the effect and possible mechanism of fat transplantation in the treatment of allergic contact dermatitis (allergic contact dermatitisus) model mice induced by 2-di-4-dinitrochlorobenzene (DNCBs). Methods A model of ACD mice was established by DNCB. The mice were randomly divided into two groups: group A (fat transplantation group), group B (sham operation group), group D (drug treatment group) and group D (natural repair group). The therapeutic effect of fat transplantation on ACD mice was observed. The histopathological changes of skin were observed by hematoxylin and eosin staining, and the expression of interleukin-10 (IL-10), interleukin-17 (IL-17) and interferon- 纬 (IFN- 纬) were detected by real-time quantitative polymerase chain reaction (Q RT-PCR). Results in group A, the thickness of epidermis returned to normal. RT-PCR analysis showed that the levels of IL-10 and IL-17 mRNA in skin tissue of group A C were lower than those of group D (P0.05), but there was no significant difference between group B and group D (P0.05). Conclusion Fat transplantation can reduce the expression of IL-10 and IL-17, promote the production of IFN- 纬 and alleviate the inflammation of lesions.
