涤痰汤对动脉粥样硬化兔模型炎性细胞因子的相关研究

【摘要】：目的:通过对动脉粥样硬化新西兰兔模型灌胃涤痰汤水煎液,检验AS兔的病理组织改变、细胞、分子生物水平,研究涤痰汤水煎液对AS兔炎性因子的影响。方法:48只雄性新西兰白兔按随机数字表法,分为6组,每组8只,即空白组、AS模型组、涤痰汤低、中、高剂量组和阿托伐他汀钙组。12周后,用全自动生化分析仪测定血清中Tc、TG、LDL-c、HDL-c、SOD和MDA的含量;ELISA法测定血清中TNF-α、IL-6、ICAM-1、VCAM-1;于主动脉弓处取直径3 mm的组织用Western blot检测作主动脉NF-κB、E-selectin蛋白表达、用荧光定量PCR检测TNF-a和IL-6mRNA表达及用免疫组化法检测ICAM-1、VCAM-1表达检测。结果:涤痰汤能降低AS兔血清TC、TG、HDL-C、LDL-C、MDA、TNF-α、IL-6、ICAM-1、VCAM-1,增高SOD水平,降低AS兔主动脉NF-κB、E-selectin蛋白的表达,并降低As兔主动脉TNF-α、IL-6 mRNA的表达,且呈剂量依赖性,与模型组比较,差异具有显著性(P0.01或P0.05)。结论:动脉粥样硬化的发生、发展与血脂异常、炎症反应的关系密切;涤痰汤具有调血脂作用,同时,能提高抗氧化应激能力,具有降低致动脉粥样硬化炎性细胞因子的作用。
[Abstract]:Objective: to study the effect of Ditan decoction on inflammatory factors of as rabbits by perfusing it into stomach and examining the pathological changes, cellular and molecular biological level of as rabbits. Methods A total of 48 male New Zealand white rabbits were randomly divided into 6 groups with 8 rabbits in each group. The rats were divided into two groups: control group (n = 8), Ditan decoction group (n = 8), low, medium and high dose group and Atto vastatin calcium group (n = 12). The contents of SOD and MDA in serum were determined by automatic biochemistry analyzer. The expression of NF- 魏 BE-selectin in aorta was detected by Western blot, and the expression of TNF- 魏 BE-selectin in aortic arch was detected by Western blot, and the expression of TNF- 伪 IL-6, ICAM-1 and VCAM-1 in serum was determined by enzyme-linked immunosorbent assay (Elisa), and the expression of NF- 魏 BE-selectin in aorta was detected by Western blot. The expression of TNF-a and IL-6 mRNA was detected by fluorescence quantitative PCR and the expression of ICAM-1 and VCAM-1 was detected by immunohistochemistry. Results: Ditan decoction could decrease the level of SOD, decrease the expression of NF- 魏 B and E-selectin and decrease the expression of TNF- 伪 IL-6 mRNA in as rabbit aorta in a dose-dependent manner. The difference was significant compared with the model group (P0.01 or P0.05). Conclusion: the occurrence and development of atherosclerosis are closely related to abnormal blood lipids and inflammatory reaction. Ditan decoction has the effect of regulating blood lipids at the same time it can improve the ability of antioxidant stress and reduce the inflammatory cytokines of atherosclerosis.
