MRSA大鼠感染性心内膜炎模型的建立
[Abstract]:Objective to establish the rat model of endocarditis infected with MRSA by using ATCC43300 standard strain. Methods the rats were intubated to the left ventricle through the right common carotid artery. After 24 hours of operation, the rats were injected into the tail vein with the amount of ATCC43300 bacteria only infected for 10 ~ 3 ~ 10 ~ (4) and 10 ~ (5) CFU/. The rats were killed on the 5th and 7th day after infection, and the cardiac neoplasm, spleen and kidney were collected for colony counting. At the same time, the non-operative infection group and the operative non-infection group were used as the control group. Results after 1 day of infection, there were vegetations in the valve of rats with cardiac catheterization, and even wall vegetations were formed in severe cases, and with the increase of the concentration of infected bacteria. The infection degree of endocarditis was increased. The infection rate of 10 ~ 4 CFU/ only group was 10 ~ 4 CFU/. The logarithmic values of bacterial count of neoplasm were 7.04 卤1.389.32 卤1.22 ~ 9.78 卤1.03.The bacterial endocarditis infection was the most serious after 3 days of infection. The maximum logarithmic value of bacterial count was 10.03 卤0.71, and the infection degree decreased with the infection amount of 10 ~ 3 CFU/ and 10 ~ 4 CFU/, respectively, to 6.87 卤0.63 and 8.23 卤1.05.The infection tendency of spleen and kidney was approximately the same as that of heart. But the degree of infection is lower than that of the heart. No neoplasm was found in the surgical non-infection group and non-operative infection group. Conclusion the model established by this method has the characteristics of strong maneuverability, simplicity, stability, good reproducibility and high success rate, and is more suitable for the study of antimicrobial mechanism, new drug screening and evaluation, and endocarditis related diseases.
