AB血清对人体外终末期红系分化进程的影响

【摘要】:目的:探究体外诱导造血干细胞向红系分化过程中AB血清对终末期红系分化进程的影响。方法:从脐带血中用免疫磁珠法分离提取CD34+细胞,体外诱导其向红系分化。根据培养体系中是否添加AB血清,设置实验组为添加AB血清组,对照组为不添加AB血清组。通过检测细胞表面红系分化标志分子糖化血红蛋白A(glycophorin A,GPA)、人红细胞阴离子交换蛋白Band3及整合素α4(α4-integrin)的表达反映终末红系分化进程,用流式细胞术检测红系细胞终末分化脱核情况。结果:与对照组相比,实验组GPA阳性细胞百分率较高,Band3表达较高,而α4-integrin表达较低(P0.05)。实验组细胞脱核百分率明显高于对照组(P0.05)。结论:在体外红系发育培养体系中,AB血清对终末期红系分化进程有一定的促分化、促脱核作用。
[Abstract]:Aim: to investigate the effect of AB serum on the process of erythroid differentiation induced by hematopoietic stem cells in vitro. Methods: CD34 cells were isolated from umbilical cord blood by immunomagnetic beads and induced to differentiate into erythrocytes in vitro. According to whether the AB serum was added in the culture system, the experimental group was added AB serum group, and the control group was no AB serum group. The expression of human erythrocyte anion exchange protein (Band3) and integrin 伪 4 (伪 4-integrin) were detected to reflect the process of terminal erythroid differentiation. Flow cytometry was used to detect the denucleation of erythrocyte terminal differentiation. Results: compared with the control group, the percentage of GPA positive cells in the experimental group was higher than that in the control group, while the expression of 伪 4-integrin was lower (P0.05). The percentage of cell denucleation in the experimental group was significantly higher than that in the control group (P0.05). Conclusion: in vitro erythroid development culture system, the serum Asab can promote the differentiation and denucleation of the end-stage erythroid.
