帕金森病模型小鼠脾悬液中辅助性T细胞17和髓系抑制性细胞数量的变化及其意义
[Abstract]:Objective to investigate the changes and significance of helper T cell 17 (Th17) and myeloid inhibitory cell (MDSC) (MDSC) in spleen suspension of (PD) mice with Parkinson's disease. Methods 40 C57BL/6 mice were randomly divided into PD group (group A), gemcitabine group (group B), gemcitabine group (group C) and control group (group D). Group B and group C received intraperitoneal injection of gemcitabine (120 mg/kg) intraperitoneally on the 7th day after 7 days of irradiation with normal saline of the same volume. The number of tyrosine hydroxylase (TH) positive cells in substantia nigra and the percentage of Th17 and MDSC cells in spleen suspension were measured after behavioral experiment on the 8th day. Results compared with those before modeling, the turning time, climbing time and suspending time of group A and B mice were significantly prolonged (P0.05). Compared with group C and group D, the time of turning head and climbing pole in group A and group B were significantly prolonged after modeling. The suspension time was significantly shortened (P0.05). The number of TH positive cells in the substantia nigra of group A was significantly lower than that in group C and group D (P0.05). The percentage of Th17,MDSC cells in splenic suspension of group A was significantly higher than that of group C and group D (P0.05). The percentage of Th17 cells in splenic suspension of group B was significantly higher than that in group A (P0.05). Pearson correlation analysis showed that there was no correlation between the percentage of Th17 and MDSC in group A and group B (r-0.319r0.04, P0.05). Conclusion the increased number of Th17,MDSC in PD model mice may be involved in the development of PD. MDSC may play an immunoregulatory role in inhibiting Th17 amplification.
