忍冬藤颗粒对急性痛风性关节炎模型大鼠形态学及关节软骨IL-1β的影响

【摘要】:目的:研究忍冬藤颗粒治疗急性痛风性关节炎模型大鼠抗炎消肿止痛的效果并对其机理进行探讨。方法:给予大鼠乙胺丁醇(189mg/kg)和腺嘌呤(1.26g/kg)灌胃,每天1次连续3周诱导高尿酸血症模型,然后在此基础上以大鼠的右踝关节外侧后方为穿刺点,针尖斜向前上方与胫骨成45°角刺入关节腔,每只注射0.2ml尿酸钠溶液,连续3天。同时各干预组大鼠分别灌胃忍冬藤颗粒23.8g/kg、11.9g/kg、5.95g/kg,别嘌呤醇0.455g/kg(第3周开始,连续1周),秋水仙碱18.9mg/kg(第4周开始.连续1周)进行干预。实验结束后,观察忍冬藤颗粒对模型大鼠关节肿胀、步态、炎症的影响,检测大鼠关节软骨中的IL-1β的表达水平。结果:与空白组比较,模型组大鼠关节肿胀增加(6.20±0.19),关节炎症(5.25±1.04)、跛行步态评分(5.25±1.04)增高,关节软骨IL-1β表达水平上升(12.33±1.36);与模型组比较,药物干预24h、48h、72h后,忍冬藤颗粒各组大鼠关节肿胀度周径明显减小,其中忍冬藤颗粒11.9g/kg剂量组对关节肿胀度的疗效更好(4.51±0.25);与模型组比较,给药后72h后,秋水仙碱(18.9mg/kg)+别嘌呤醇(0.455g/kg)组可以明显降低关节炎症程度,给药48、72h后,忍冬藤各剂量组可以明显降低关节炎症评分,其中忍冬藤颗粒11.9g/kg剂量组降低炎症评分效果更优(1.00±1.07);与模型组比较,给药后24h、48h、72h后,忍冬藤颗粒各剂量组可以明显降低步态评分(1.00±1.07);与模型组比较,秋水仙碱(18.9mg/kg)+别嘌呤醇(0.455g/kg)、忍冬藤颗粒各剂量组大鼠血清IL-1β表达明显降低,其中忍冬藤颗粒11.9g/kg(6.16±0.70)、23.8g/kg(6.96±0.79)剂量组IL-1β表达降低效果最优。结论:忍冬藤颗粒可以起到抗炎消肿止痛的功效,对急性痛风性关节炎模型大鼠有着明显的治疗作用,其作用机制可能是抑制炎症关节IL-1β致炎因子的生成而实现的。
[Abstract]:Objective: to study the effect and mechanism of Lonicera japonicus granule in treating acute gouty arthritis model rats. Methods: rats were given ethambutanol (189mg/kg) and adenine (1.26g/kg) intragastrically to induce hyperuricemia model once a day for 3 weeks. A 45 掳angle between the tip oblique and the tibia was inserted into the articular cavity. Each mouse was injected with sodium 0.2ml uric acid for 3 days. At the same time, the rats in each intervention group were treated by intragastric administration of Lonicera japonicus granules 23.8g / kg 11.9g / kg, allopurinol 0.455g/kg 5.95g / kg, and colchicine 18.9mg/kg at the beginning of week 3, respectively, and the rats were given colchicine 18.9mg/kg (from week 4). Intervention was carried out for one week in a row. At the end of the experiment, the effects of Lonicera japonicus granule on joint swelling, gait and inflammation were observed, and the expression level of IL-1 尾 in articular cartilage of rats was detected. Results: compared with the blank group, the joint swelling increased (6.20 卤0.19), arthritis (5.25 卤1.04), limping gait score (5.25 卤1.04) and articular cartilage IL-1 尾 expression increased (12.33 卤1.36) in the model group. The radius of swelling degree of joint decreased obviously in each group, and the effect of 11.9g/kg group was better (4.51 卤0.25), compared with the model group, 72 hours after administration, the effect was better than that in the model group, and compared with the model group, after 72 hours of administration, the effect of 11.9g/kg on the degree of joint swelling was better than that in the model group. Colchicine (18.9mg/kg) allopurinol (0.455g/kg) group could significantly reduce the degree of arthritis. Compared with the model group, each dose group of Lonicera Lonicera granule at 48 hours and 72 hours after administration significantly decreased the gait score (1.00 卤1.07), compared with the model group, and compared with the model group, each dose group of Lonicera Lonicera granule decreased the score of gait obviously (1.00 卤1.07), compared with the model group. The expression of serum IL-1 尾 in the colchicine (18.9mg/kg) allopurinol (0.455g/kg) and Lonicera japonica granule groups was significantly decreased, and the decrease of IL-1 尾 expression was the best in the group of 11.9g/kg (6.16 卤0.70) and 23.8g / kg (6.96 卤0.79). Conclusion: Lonicera japonicus granule can inhibit inflammation, detumescence and pain, and has obvious therapeutic effect on acute gouty arthritis rats, and its mechanism may be to inhibit the production of IL-1 尾 inflammatory factor in inflammatory joints.
