谷氨酸和肽聚糖在促进细菌间纳米管形成中作用的研究

【摘要】:目的对细菌间纳米管形成机制及组成成分进行探索,为预防细菌耐药提供潜在靶标。方法利用扫描电子显微镜观察细菌间纳米管的形成,用表面解吸常压化学电离质谱(SDAPCI-MS)技术和酶联免疫吸附实验(ELISA)分别检测细菌间纳米管形成前后谷氨酸(glutamic,Glu)和肽聚糖(peptidoglycan,PGN)的浓度。结果相同菌量的金黄色葡萄球菌和大肠埃希菌在纳米管形成后PGN含量明显增多,且它们在纳米管形成后消耗了更多的Glu。结论 PGN是纳米管组成成分,Glu是促进细菌间纳米管形成的主要能量来源。
[Abstract]:Objective to explore the formation mechanism and components of bacterial nanotubes and to provide a potential target for the prevention of bacterial drug resistance. Methods the formation of interbacterial nanotubes was observed by scanning electron microscope (SEM). The concentrations of glutamate (glutamic,Glu) and peptidoglycan (peptidoglycan,PGN) before and after the formation of interbacterial nanotubes were detected by surface desorption chemical ionization mass spectrometry (SDAPCI-MS) and enzyme linked immunosorbent assay (ELISA). Results the PGN content of Staphylococcus aureus and Escherichia coli increased significantly after nanotube formation, and they consumed more Glu. after nanotube formation. Conclusion PGN is the main energy source to promote the formation of nanotubes.
