培元解郁方对几种抑郁模型小鼠的抗抑郁作用及机制研究
[Abstract]:Objective to study the antidepressant effect and mechanism of Peiyuan Jieyu prescription. Methods mice model of depression was established by intraperitoneal injection of reserpine 2.5 mg/kg, depression model of mice under acute stress was established by forced swimming, depression model of mice under chronic stress was established by long-term (14 d) restraint plus noise. To observe the effect of Peiyuan Jieyu recipe on body temperature, blepharoptosis and loop rate of reserpine depression model mice, and on the immobility time of forced swimming in acute stress model and chronic stress model mice. The contents of 5-hydroxytryptamine (5-HT), norepinephrine (NA) and indoleamine 3-dioxygenase (IDO) in brain of acute stress model mice were detected by enzyme-linked immunosorbent assay (ELISA). Fluorescence quantitative PCR was used to detect the expression of IDO mRNA in the brain of chronic emergency model mice. Results Peiyuan Jieyu prescription could antagonize the hypothermia of reserpine depression model mice (P0.01). In addition, high dose could also improve the condition of eyelid droop and motor failure induced by reserpine (P0.05), while the middle dose group could improve the condition of motility (P0.05). Peiyuan Jieyu recipe could shorten the immobility time (P0.05U P0.01) and significantly increase the content of 5-HTNNA in the brain (P0.05U P0.01) in the low dose group. Peiyuan Jieyu prescription could shorten the immobility time of forced swimming in chronic stress model mice, increase the content of 5-HT in brain tissue, decrease the content of IDO and up-regulate the expression of IDO mRNA (P0.05). Conclusion Peiyuan Jieyu prescription can ameliorate the depression symptoms of mice caused by reserpine, forced swimming and chronic stress. The mechanism may be related to the increase of monoamine neurotransmitter 5-HT na, the inhibition of IDO activity and the down-regulation of IDO mRNA expression in the brain of depression model mice.
