人胰岛素放射免疫分析试剂盒测定小型猪胰岛素的可行性研究

【摘要】:目的探讨人特异的胰岛素放射免疫分析试剂盒(Ins-RIA)检测小型猪血清胰岛素水平的可行性。方法采集219份巴马小型猪血清标本,分别采用人Ins-RIA和猪特异的胰岛素ELISA试剂盒(Ins-ELISA)测定血清胰岛素水平。分析两种方法检测猪胰岛素水平的相关性,随机抽取80%(175份)的数据进行直线回归分析,以Ins-ELISA为因变量,Ins-RIA为自变量,建立回归方程,再用剩余20%(44份)的数据对回归方程进行回代验证。结果在巴马小型猪,猪Ins-ELISA检测值低于人Ins-RIA检测值,分别为(15.32±15.50)μIU/mL及(32.31±21.74)μIU/mL,差异有统计学意义(P0.000 1);预测猪Ins-ELISA检测胰岛素的回归方程为InsELISA(μIU/mL)=-7.29+0.70×Ins-RIA(μIU/mL)(R~2=0.94);在回代验证中,Ins-ELISA的预测值和实际检测值之间的平均差异为17.18%,预测值与实际测量值之间的吻合程度较好(P0.05)。结论巴马小型猪ELISA法检测的血清胰岛素水平可以用人胰岛素RIA法检测的血清胰岛素水平来进行估计。
[Abstract]:Objective to investigate the feasibility of human specific insulin radioimmunoassay kit (Ins-RIA) for detection of serum insulin in miniature pigs. Methods Human Ins-RIA and porcine specific insulin ELISA kit (Ins-ELISA) were used to detect serum insulin level in 219 Bama miniature pig serum samples. By analyzing the correlation between the two methods to detect the insulin level in pigs, 80% (175 samples) of the data were randomly sampled for linear regression analysis. The regression equation was established with Ins-ELISA as the dependent variable and Ins-RIA as the independent variable. The regression equation was verified with the data of the remaining 20% (44 copies). Results in Bama mini-pigs, the detection value of pig Ins-ELISA was lower than that of human Ins-RIA. There were significant differences between (15.32 卤15.50) 渭 IU/mL and (32.31 卤21.74) 渭 IU/mL, respectively (P0.0001), the regression equation of predicting insulin detection by Ins-ELISA was InsELISA (渭 IU/mL) -7.29 0.70 脳 Ins-RIA (渭 IU/mL) (RP2O0.94), and the average difference between the predicted value of Ins-ELISA and the actual value of detection was 17.18% in the back generation verification, and the regression equation was InsELISA (渭 IU/mL) -7.29 0.70 脳 Ins-RIA (渭 IU/mL), and the average difference between the predicted value and the actual measured value was 17.18. The degree of anastomosis was good (P0.05). Conclusion the serum insulin level measured by ELISA method in Bama miniature pigs can be estimated by the serum insulin level measured by human insulin RIA method.
