基于活体成像技术的裸鼠卵巢癌休眠模型的建立

【摘要】:目的建立基于活体成像的裸鼠卵巢癌休眠模型并利用小动物活体成像技术验证休眠细胞的存在。方法采用慢病毒载体对人卵巢癌skov-3细胞稳定转染萤火虫荧光素酶基因(firefly luciferase gene),经筛选获得稳定表达荧光素酶基因的人卵巢癌细胞株(skov-3/luc)。将skov-3/luc细胞接种至裸小鼠右侧腋部皮下,成瘤后观测荧光并做肿瘤切除手术,术后的裸鼠再常规喂养8周后选取接种肿瘤处肉眼未见肿瘤组织生长的裸鼠进行活体成像观察,显示接种处肿瘤细胞荧光阳性者,即认为卵巢癌裸鼠休眠模型建立成功。结果筛选到了稳定表达Luc的skov-3/luc细胞株;成功构建了基于活体成像的裸鼠卵巢癌休眠模型,并利用小动物活体成像技术验证了裸鼠体内卵巢癌休眠细胞的存在。结论利用活体成像技术构建的裸鼠卵巢癌休眠模型为研究肿瘤休眠的机制及诱导和维持肿瘤细胞休眠的方法提供了理想的实验动物模型。
[Abstract]:Objective to establish a dormant model of ovarian cancer in nude mice based on in vivo imaging and to verify the existence of dormant cells by small animal imaging in vivo. Methods Human ovarian cancer cell line (skov-3/luc) with stable expression of luciferase gene was obtained by using lentivirus vector and stable transfection of luciferase gene (firefly luciferase gene), into human ovarian cancer skov-3 cells. Skov-3/luc cells were inoculated into the right axillary subcutaneous of nude mice. Fluorescence was observed after tumorigenesis and tumor resection was performed. After 8 weeks of routine feeding, nude mice inoculated with the tumor were selected to observe the growth of tumor tissue in vivo. Those who showed positive fluorescence of tumor cells at inoculation were considered to be successful in establishing dormancy model of ovarian cancer in nude mice. Results the skov-3/luc cell lines expressing Luc stably were screened, and the dormancy model of ovarian cancer in nude mice was successfully constructed based on in vivo imaging, and the existence of ovarian cancer dormant cells in nude mice was verified by small animal imaging in vivo.
