左归丸对肾阴虚模型大鼠神经-内分泌-免疫功能的影响

【摘要】:目的:探讨左归丸对肾阴虚模型大鼠神经-内分泌-免疫功能的作用机制。方法:按随机数字将60只清洁级健康雄性Wistar大鼠分均为正常组(同体积生理盐水),模型组(同体积生理盐水),预防组(与造模同时,1 g·kg~(-1)左归丸)及左归丸低剂量组(造模后,0.5 g·kg~(-1)左归丸),中剂量组(造模后,1 g·kg~(-1)左归丸),高剂量组(造模后,2 g·kg~(-1)左归丸),以甲状腺片灌胃制备肾阴虚模型;所有组均连续灌胃21 d。采集脾、胸腺、尿液和血液样本,计算脏器指数;采用酶联免疫吸附法(ELISA)检测促肾上腺皮质素释放激素(CRH),促肾上腺皮质激素(ACTH),皮质醇(CORT),三碘甲状腺原氨酸(T3),四碘甲状腺原氨酸(T4),甲状腺刺激激素(TSH),环磷酸腺苷(c AMP),环磷酸鸟苷(c GMP),17-羟皮质类固醇(17-OHCS),白细胞介素-6(IL-6),干扰素-γ(IFN-γ)水平。结果:与正常组比较,模型组的T3,T4,c AMP,c GMP,IL-6,IFN-γ明显升高,TSH,c AMP/c GMP,脾指数,胸腺指数降低,差异显著(P0.05,P0.01),预防组各指标无明显变化。与模型组比较,左归丸各剂量组的T3,T4,c AMP,c GMP,IL-6,IFN-γ明显降低,TSH,c AMP/c GMP,脾指数、胸腺指数明显升高,差异显著(P0.05,P0.01);预防组T3,T4,CRH,ACTH,CORT,c AMP,c GMP,17-OHCS,脾指数,胸腺指数明显升高(P0.01),TSH,c AMP/c GMP,IL-6,IFN-γ明显降低(P0.01)。与预防组比较,左归丸各剂量组的T3,CRH,ACTH较高(P0.05,P0.01)。结论:左归丸能调节甲亢型肾阴虚大鼠下丘脑-垂体-肾上腺轴和下丘脑-垂体-甲状腺轴功能,改善免疫功能,这可能是干预甲亢型肾阴虚作用的机制之一。
[Abstract]:Objective: to investigate the mechanism of Zuogui Pill on neuroendocrine-immune function in rats with kidney yin deficiency. Methods: sixty clean grade healthy male Wistar rats were randomly divided into normal group (normal saline of the same volume), model group (same volume of saline), prevention group (1 g kg~ (-1) Zuogui pill) and low dose of Zuogui pill. The model group (0.5 g kg~ (-1) Zuo Gui Wan), the middle dose group (1 g kg~ (-1) Zuo Gui Pill) and the high dose group (2 g kg~ (-1) Zuo Gui Pill) were perfused with thyroid tablets to establish the model of kidney yin deficiency. All the groups were perfused continuously for 21 days. Spleen thymus urine and blood samples were collected to calculate the visceral index. Enzyme linked immunosorbent assay (ELISA) for the detection of adrenocorticotropin releasing hormone (CRH), adrenocorticotropic hormone (ACTH), cortisol, (CORT), triiodothyronine (T3), tetraiodothyronine (T4), and thyroid stimulating hormone (TSH), cyclic adenosine phosphate (c AMP),) The levels of (c GMP), 17-hydroxycorticosteroid (17-OHCS), interleukin-6 (IL-6) and interferon- 纬 (IFN- 纬) were measured. Results: compared with the normal group, the T _ (3) T _ (4) C AMP,c GMP,IL-6,IFN- 纬 in the model group was significantly higher than that in the control group, but the spleen index and thymus index of the model group were decreased significantly (P 0.05 ~ (0.01), but no significant change was found in the prevention group. Compared with the model group, the T _ (3) T _ (4) C AMP,c GMP,IL-6,IFN- 纬, TSH,c AMP/c GMP, spleen index and thymus index in each dose group were significantly lower than those in the model group (P < 0.05), while in the prevention group, the spleen index, thymus index and TSH,c AMP/c GMP,IL-6,IFN- 纬 were significantly increased (P ~ (0.01) and significantly decreased (P ~ (0.01). Compared with the prevention group, the T _ 3 CRH ACTH of each dose group was higher than that of the prevention group (P 0.05? P 0.01). Conclusion: Zuogui Pill can regulate the function of hypothalamus-pituitary-adrenal axis and hypothalamus-pituitary-thyroid axis and improve immune function in hyperthyroidism rats.
