木瓜蛋白酶与石膏制动建立兔膝骨关节炎模型的比较

【摘要】:目的比较木瓜蛋白酶关节腔注射与关节石膏伸直位制动建立兔膝骨关节炎模型的效果。方法将21只实验兔采用随机区组分组法分为正常组、木瓜蛋白酶组及石膏组,每组7只。木瓜蛋白酶组予6%木瓜蛋白酶生理盐水溶液于实验开始第1、4、7天行右膝关节腔注射,注射结束4周后造模完成;石膏组予膝关节伸直位固定6周;记录实验期间动物不良情况发生率,造模完成后处死实验兔,取软骨标本进行大体观察、Pelletier评分、Mankin's评分,免疫组化测定软骨Ⅱ型胶原蛋白含量,ELISA法检测血清和关节液中肿瘤坏死因子-α(TNF-α)、软骨寡聚基质蛋白(COMP)以及基质金属蛋白酶-3(MMP-3)的含量。结果大体观察显示木瓜蛋白酶组与石膏组均出现关节肿胀、关节间隙变窄、软骨缺损、滑膜增生硬化、炎症改变、骨赘形成;Pelletier评分、Mankin's评分、Ⅱ型胶原蛋白含量、血清和关节液中TNF-α、COMP以及MMP-3的含量,木瓜蛋白酶组、石膏组与正常组比较,差异均有统计学意义(P0.05);木瓜蛋白酶组与石膏组比较,上述指标差异均无统计学意义(P0.05)。木瓜蛋白酶组动物不良情况发生率明显低于石膏组,差异有统计学意义(P0.01)。结论木瓜蛋白酶关节腔注射与石膏关节伸直位制动均能成功建立膝骨关节炎模型,但木瓜蛋白酶关节腔注射法更为快捷、简便,并发情况少,可行性好。
[Abstract]:Objective to compare the effect of papain injection and plaster immobilization on rabbit knee osteoarthritis model. Methods 21 rabbits were randomly divided into normal group, papain group and gypsum group with 7 rabbits in each group. In the papain group, 6% papain saline solution was injected into the right knee joint on the 1st and 4th day of the experiment, and the model was made after 4 weeks of injection, while the gypsum group was fixed in the straight position of the knee joint for 6 weeks.
