HIF-1α在失血性动物心肌中的表达

【摘要】:目的:探讨不同程度失血大鼠心肌细胞中缺氧诱导因子-1ɑ(hypoxia-inducible factor-1ɑ)蛋白和mRNA的表达和意义。 方法:选用清洁级SD大鼠,建立不同程度的失血模型,随机分为对照组(CON),失血10%(10-BL),失血20%(20-BL),失血30%(30-BL),利用Powerlab八道生理记录仪监测心功能指标,采用RT-PCR及Western Blot检测HIF-1ɑm RNA及蛋白在失血后30min,1h大鼠心肌组织中的表达情况,并用免疫组化染色检测HIF-1ɑ在心肌细胞中的分布。 结果:不同程度失血后,心功能均降低,1h比30min降低明显(P0.05);RT-PCR半定量结果显示,与CON组相比,10-BL、20-BL、30-BL组HIF-1ɑmRNA表达逐渐增高(P0.05);免疫组化结果表明,HIF-1α主要分布在心肌细胞细胞质中,在细胞核内也有分布。 结论:随失血程度增大,时间延长,HIF-1α表达增加,HIF-1α可能参与失血导致的低氧状态下心肌代谢的适应性调节。 目的:探讨不同程度失血后猪心肌细胞中缺氧诱导因子-1ɑ(hypoxia-inducible factor-1ɑ)蛋白和mRNA的表达和意义。 方法:选用小白猪,建立不同程度的失血模型,随机分为对照组(CON),失血10%(10-BL),失血20%(20-BL),失血30%(30-BL),利用Powerlab八道生理记录仪监测心功能指标,采用RT-PCR及Western Blot检测HIF-1ɑmRNA及蛋白在失血后1h小白猪心肌组织中的表达情况,并用免疫组化染色检测HIF-1ɑ在心肌细胞中的分布。 结果:不同程度失血后,心功能指标降低;RT-PCR半定量结果显示,与CON组相比,10-BL、20-BL、30-BL组HIF-1ɑmRNA表达逐渐增高(P0.05);免疫组化结果表明,HIF-1α主要分布在心肌细胞细胞质中,在细胞核内也有分布。 结论:失血程度增大,时间延长,HIF-1α表达增加,HIF-1α可能参与失血导致的低氧状态下心肌代谢的适应性调节。
[Abstract]:Aim: to investigate the expression and significance of hypoxia inducible factor-1 (hypoxia-inducible factor-1) protein and mRNA in cardiac myocytes of rats with different degrees of blood loss. Methods: blood loss models were established in clean grade SD rats. The rats were randomly divided into three groups: (CON), 10% (10-BL), bleeding 20% (20-BL), and bleeding 30% (30-BL). The cardiac function was monitored by Powerlab eight-channel physiological recorder, and the expression of HIF-1 m RNA and protein was detected by RT-PCR and Western Blot in the myocardium of rats 30 min after blood loss. The distribution of HIF-1 in cardiomyocytes was detected by immunohistochemical staining. Results: compared with CON group, the expression of HIF-1 mRNA in 10-BL20-BL 30-BL group was increased gradually (P0.05), and the results of RT-PCR semi-quantitative analysis showed that the cardiac function decreased significantly at 1 hour after different degree of blood loss (P 0.05), and the expression of HIF-1 mRNA in 10-BL20-BL 30-BL group was significantly higher than that in CON group (P0.05). Immunohistochemical results showed that HIF-1 伪 was mainly distributed in the cytoplasm of myocardium and also in the nucleus. Conclusion: with the increase of blood loss, the expression of HIF-1 伪 is increased. HIF-1 伪 may be involved in the adaptive regulation of myocardial metabolism in hypoxic state induced by blood loss. Methods: a blood loss model was established in small white pigs. The model was randomly divided into three groups: 10% (10-BL), 20% (20-BL), and 30% (30-BL) of (CON), in control group. The cardiac function was monitored by Powerlab eight-channel physiological recorder. The expression of HIF-1 mRNA and protein was detected by RT-PCR and Western Blot, and the distribution of HIF-1 in cardiac myocytes was detected by immunohistochemical staining. Results: after different degree of blood loss, cardiac function index decreased, RT-PCR semi-quantitative results showed that compared with CON group, the expression of HIF-1 mRNA increased gradually in 10-BL20-BL 30-BL group (P0.05). Immunohistochemical results showed that HIF-1 伪 was mainly distributed in the cytoplasm of myocardium and also in the nucleus. Conclusion: the degree of blood loss is increased, the time is prolonged and the expression of HIF-1 伪 is increased. HIF-1 伪 may be involved in the adaptive regulation of myocardial metabolism in hypoxic state induced by blood loss.
