人与猕猴不同组织多能性干细胞相关基因和标志抗原的表达分析

【摘要】:近来研究表明脐带、脐带血、羊水以及出生后外周血液中存在具有多分化潜能的干细胞,但研究结果不尽一致;Oct-4、Nanog、Sox-2、Rex-1、SSEAs作为胚胎干细胞的标志分子,常用于评价干细胞的多潜能性。本研究采用半定量RT-PCR法分析多能性干细胞相关基因Oct-4、Nanog、Sox-2以及Rex-1在人源脐带、脐带血、羊水组织以及猕猴外周血液中的表达情况,并对上述基因的扩增片段进行同源分析;同时用SP免疫组织化学法分析Oct-4、Nanog、Sox-2、SSEA-1、SSEA-3、SSEA-4在人源脐带组织中的表达和分布情况,其结果如下: 1、Oct-4、Nanog、Sox-2以及Rex-1基因在人源脐带、脐带血、羊水组织中均稳定表达(阳性率≥80%);在上述组织中,Oct-4、Nanog, Sox-2以及Rex-1基因的表达水平存在差异,其在脐带和羊水中表达水平差异不显著(P0.05),但二者都显著高于脐带血(P0.05) 2、在不同生长阶段健康猕猴外周血液中Oct-4、Nanog和Sox-2基因均呈阳性表达(阳性表达率≥80%);Oct-4、Nanog和Sox-2基因在猕猴外周血液中的表达水平与年龄呈负相关,但差异不显著(P0.05)。 3、将人源脐带、脐带血、羊水、猕猴外周血组织中扩增所得Oct-4、Nanog、Sox-2基因序列,与人、猕猴、猪、牛、小鼠、褐家鼠、狗、兔等部分物种上述基因cDNA序列同源性比较,结果显示,人与猕猴、兔、猪、牛有较高同源性(75%-95.9%),与小鼠和褐家鼠的同源性60%以上;上述基因序列生物进化分析的结果显示,克隆所得目的基因序列与人、猕猴相同基因的序列亲缘关系最近,三者构成亲缘关系最近的一个分支;再与猪、牛和狗的该基因构成一个较大的分支,其次亲缘关系依次为兔、大鼠和小鼠等。 4、在人源脐带组织中,均检测到表达Oct-4、Nanog、Sox-2、SSEA-1、SSEA-3、SSEA-4等标记分子的阳性细胞,阳性(阳性程度大于或等于(++))表达率在75%以上,且各标记分子阳性表达率差异不显著(P0.05),这些细胞主要分布于脐带组织的华尔通胶和脐血管周围,但血管内皮附近分布较少。

As a marker of embryonic stem cells, Oct-4,Nanog,Sox-2,Rex-1,SSEAs is often used to evaluate the pluripotency of stem cells. The expression of Oct-4,Nanog,Sox-2 and Rex-1 in human umbilical cord, umbilical cord blood, amniotic fluid and peripheral blood of rhesus monkey were analyzed by semi-quantitative RT-PCR. The amplified fragments of the above genes were analyzed by homology analysis.
