电针刺激对脊髓挫伤模型大鼠神经细胞凋亡的影响

【摘要】:背景:实验研究表明,电针对于脊髓损伤的功能恢复具有促进作用,可以降低疼痛,提高急性脊髓损伤的术后疗效。目的:观察脊髓挫伤造模后电针刺激干预对损伤区细胞凋亡的影响,并分析其对脊髓挫伤大鼠神经功能保护的可能作用机制。方法:成年雌性SD大鼠66只,采用随机区组法分为假手术组20只和大鼠急性脊髓挫伤造模成功40只,造模成功大鼠再随机分为脊髓挫伤组、电针刺激组。分别于造模前、造模后1 d、3 d、1周、2周、3周、4周通过BBB评分、斜板试验进行运动功能评定。造模后3 d,通过TUNEL法检测各实验组大鼠脊髓挫伤区神经细胞的凋亡情况;RT-PCR、Western blot检测脊髓挫伤区周围bax、bcl-2、caspase-3基因转录和蛋白的表达;苏木精-伊红染色观察脊髓挫伤区病理组织形态学变化;HRP示踪观察神经纤维的再生情况。结果与结论:1电针刺激组2项运动功能评分在治疗后2周各个时间点均较脊髓挫伤组显著增加(P0.05)。2造模后3 d经脊髓电针刺激组细胞凋亡指数均明显低于脊髓挫伤组(P0.05)。3造模后72 h,与脊髓挫伤组相比经脊髓电针刺激组bax、caspase-3基因和蛋白表达显著降低(P0.05);bcl-2基因和蛋白表达显著升高(P0.05)。4造模后4周,HRP阳性神经纤维数:假手术组最多,电针刺激组次之,脊髓挫伤组最少,各组之间差异有显著性意义(P0.05)。结果提示,电针刺激治疗通过减少大鼠脊髓挫伤区神经细胞的凋亡对脊髓挫伤起到保护作用。
[Abstract]:Background: experimental studies have shown that electroacupuncture can promote the functional recovery of spinal cord injury, can reduce pain and improve the postoperative efficacy of acute spinal cord injury. Aim: to observe the effect of electroacupuncture (EA) stimulation on cell apoptosis after spinal cord contusion in rats, and to analyze the possible mechanism of its protective effect on spinal cord contusion in rats. Methods: 66 adult female SD rats were randomly divided into sham operation group (n = 20) and acute spinal cord contusion model (n = 40). The motor function was evaluated by BBB score and inclined plate test before and 1 day after model making and 1 week, 2 weeks, 3 weeks and 4 weeks after modeling. The apoptosis of neurons in the contusion area of spinal cord was detected by TUNEL method and the expression of bax,bcl-2,caspase-3 gene and protein around the contusion area of spinal cord was detected by RT-PCR,Western blot on the 3rd day after the establishment of the model. Histopathological changes in contusion of spinal cord were observed by hematoxylin-eosin staining and regeneration of nerve fibers was observed by HRP tracer. Results and conclusion: 1 the motor function scores of the electroacupuncture group were significantly higher than those of the spinal cord contusion group at every time point 2 weeks after treatment (P0.05). 2 the apoptotic index of the electroacupuncture stimulation group was significantly higher than that of the spinal cord contusion group 3 days after the treatment (P0.05). It was significantly lower than that in the spinal cord contusion group (P0.05). Compared with the spinal cord contusion group, the expression of bax,caspase-3 gene and protein decreased significantly in the spinal cord electroacupuncture group (P0.05). The expression of bcl-2 gene and protein increased significantly (P0.05). 4 the number of HRP positive nerve fibers was the most in sham operation group, followed by electroacupuncture stimulation group and spinal cord contusion group (P0.05). 4. 4 weeks after modeling, the number of HRP positive nerve fibers was the highest in sham operation group, second in electroacupuncture group, and the least in spinal cord contusion group (P0.05). The results suggest that electroacupuncture therapy can protect spinal cord contusion by reducing the apoptosis of nerve cells in spinal cord contusion area.
