痛泻要方对腹泻型肠易激综合征模型大鼠肠道5-HT受体信号传导系统的影响

【摘要】:目的:从肠道5-HT受体信号传导途径探讨痛泻要方对IBS-D模型大鼠内脏高敏感性影响的作用机制。方法:采用随机数字表法,将32只雄性SD大鼠随机分为3组:空白对照组,模型对照组,痛泻要方组。采用0.5%醋酸灌肠法来建立IBS-D内脏高敏感大鼠模型。痛泻要方组予痛泻要方灌胃(大鼠每日生药用量9 g/kg),干预2周。2周后用western blot法测定小肠和结肠黏膜组织中5-HTR1、2、3的表达。结果:模型对照组近端小肠、远端小肠、近端结肠和远端结肠的5-HT1R蛋白表达与空白对照组比较差异有统计学意义(P0.05),痛泻要方组和模型对照组相比差异有统计学意义(P0.05),与空白对照组相比差异无统计学意义(P0.05)。模型对照组近端小肠、远端小肠、近端结肠和远端结肠的5-HT2R蛋白表达与空白对照组相比差异有统计学意义(P0.05)。痛泻要方组和模型对照组相比差异无统计学意义(P0.05),与空白对照组相比差异有统计学意义(P0.05)。模型对照组近端小肠、远端小肠、近端结肠和远端结肠的5-HT3R蛋白表达与空白对照组相比差异有统计学意义(P0.05)。痛泻要方组和模型对照组相比差异有统计学意义(P0.05),与空白对照组相比远端结肠差异无统计学意义(P0.05)。结论:痛泻要方能够使IBS-D模型大鼠的内脏高敏感性降低,其机制可能是通过下调5-HT1R、5-HT3R的表达来降低模型大鼠的内脏高敏感性。
[Abstract]:Aim: to investigate the effect of Tongxie decoction on visceral hypersensitivity in IBS-D model rats by means of intestinal 5-HT receptor signal transduction pathway. Methods: 32 male SD rats were randomly divided into three groups: blank control group, model control group and Tongxie Yaofang group. The rat model of IBS-D visceral hypersensitivity was established by 0.5% acetic acid enema. Tongxieyoufang group was treated with Tongxieyoufang (9 g/kg daily crude drug dosage) for 2 weeks. After 2 weeks of intervention, the expression of 5-HTR1O2O3 in small intestine and colon mucosa was determined by western blot method. Results: the expression of 5-HT1R protein in the proximal small intestine, distal small intestine, proximal colon and distal colon in the model control group was significantly higher than that in the blank control group (P0.05). There was significant difference between Tongxieyaofang group and model control group (P0.05), but there was no significant difference compared with blank control group (P0.05). The expression of 5-HT2R protein in the proximal small intestine, distal small intestine, proximal colon and distal colon in the model control group was significantly higher than that in the blank control group (P0.05). There was no significant difference between Tongxieyaofang group and model control group (P0.05), but there was significant difference compared with blank control group (P0.05). The expression of 5-HT3R protein in the proximal small intestine, distal small intestine, proximal colon and distal colon in the model control group was significantly higher than that in the blank control group (P0.05). There was significant difference between Tongxieyangfang group and model control group (P0.05), but there was no significant difference in distal colon compared with blank control group (P0.05). Conclusion: Tongxieyangfang can reduce the visceral hypersensitivity of IBS-D rats, and its mechanism may be that it can decrease the visceral hypersensitivity of IBS-D rats by down-regulating the expression of 5-HT _ 1RN _ (5-HT _ 3R).
