银杏叶黄酮对急性脑梗死模型小鼠凋亡因子的影响

【摘要】:目的研究银杏叶黄酮对急性脑梗死模型小鼠血清低氧诱导因子-1α(HIF-1α)、肿瘤坏死因子-α(TNF-α)及半胱天冬酶-3(caspase-3)影响。方法选取昆明雌性小鼠24只,随机选取8只为假手术组,剩下小鼠进行急性脑梗死模型的构建。将模型小鼠按照体重随机分成模型组和实验组,每组8只,实验组每天腹腔注射20 mg·kg~(-1)银杏叶黄酮,假手术组和模型组同样给药方式给予0.9%NaCl,连续7 d。处死小鼠,用酶联免疫吸附法检测各组急性脑梗死模型小鼠血清中HIF-1α、TNF-α及Caspase-3的表达水平。结果假手术组、模型组、实验组小鼠血清中的TNF-α分别为(5.16±1.04),(33.02±20.34),(22.64±10.17)ng·mL~(-1);这3组小鼠血清中的HIF-1α分别为(508.94±123.18),(735.19±324.05),(626.24±212.76)ng·mL~(-1);这3组小鼠血清中的Caspase-3分别为(6.54±0.24),(12.23±3.45),(10.24±1.58)ng·mL~(-1);模型组小鼠血清中TNF-α、HIF-1α、Caspase-3表达明显高于假手术组,差异均有统计学意义(均P0.05);实验组小鼠血清中TNF-α、HIF-1α、Caspase-3表达明显低于模型组,差异均有统计学意义(均P0.05)。结论银杏叶黄酮能够明显下调急性脑梗死小鼠血清中TNF-α、Caspase-3及HIF-1α水平。
[Abstract]:Objective to study the effects of ginkgo biloba flavonoids on serum hypoxia inducible factor-1 伪 (HIF-1 伪), tumor necrosis factor- 伪 (TNF- 伪) and cysteinase 3 (caspase-3) in mice with acute cerebral infarction. Methods Twenty-four Kunming female mice were selected and 8 mice were randomly selected as sham-operated group. The remaining mice were used to construct acute cerebral infarction model. The model mice were randomly divided into model group and experimental group according to their body weight. The experimental group received 20 mg kg~ (-1) ginkgo biloba flavone intraperitoneally every day. The sham operation group and the model group were given 0.9 NaCl3 for 7 days. The expression levels of HIF-1 伪, TNF- 伪 and Caspase-3 in serum of the mice with acute cerebral infarction were detected by enzyme-linked immunosorbent assay (Elisa). Results the levels of TNF- 伪 in serum of sham operation group, model group and experimental group were (5.16 卤1.04), (33.02 卤20.34), (22.64 卤10.17) ng mL~ (-1); The HIF-1 伪 in serum of the three groups was (508.94 卤123.18), () 735.19 卤324.05), (626.24 卤212.76) ng mL~ (-1); The Caspase-3 in serum of the three groups was (6.54 卤0.24), (12.23 卤3.45), (10.24 卤1.58) ng mL~ (-1); The expression of TNF- 伪, HIF-1 伪 and Caspase-3 in serum of model group was significantly higher than that of sham operation group (P0.05). Conclusion Ginkgo biloba flavonoids can significantly decrease the levels of TNF- 伪, Caspase-3 and HIF-1 伪 in serum of mice with acute cerebral infarction.
