费城染色体阳性急性淋巴细胞白血病小鼠模型的建立和鉴定

【摘要】:目的:建立Ph~+急性淋巴细胞白血病(ALL)小鼠模型,为Ph~+ALL研究提供工具。方法:用M ig190逆转录病毒转染BABL/c小鼠骨髓细胞产生CML样小鼠,分选CML样小鼠BCR-ABL~+B细胞,输注同系小鼠建立Ph~+ALL模型;应用流式细胞术鉴定免疫表型,RT-PCR和Western blot鉴定BCR-ABL转录和蛋白表达。结果:M ig190逆转录病毒转染后小鼠迅速发生CM L样病变,输注来自CM L样小鼠的BCR-ABL~+B细胞后受鼠均发生急性淋巴细胞白血病,免疫分型显示白血病细胞CD19~+;RT-PCR和Western Blot均检测到BCR-ABL,符合Ph~+ALL的免疫表型和分子生物学特征。结论:成功开发了一种新型Ph~+ALL小鼠模型的建立方法,能够为研究Ph~+ALL提供一有力工具。
[Abstract]:Objective: to establish Ph~ acute lymphoblastic leukemia (Ph~) (ALL) mouse model and to provide a tool for the study of Ph~ ALL. Methods: M ig190 retrovirus was transfected into the bone marrow cells of BABL/c mice to produce CML like mice. BCR-ABL~ B cells of CML like mice were selected and the Ph~ ALL model was established in the same strain mice. Flow cytometry was used to identify the immunophenotype, and RT-PCR and Western blot were used to identify the transcription and protein expression of BCR-ABL. Results: M ig190 retrovirus-transfected mice developed CM L-like lesions rapidly, and acute lymphoblastic leukemia (ALL) occurred in all recipient mice after transfection of BCR-ABL~ B cells from CM L-like mice. Both RT-PCR and Western Blot detected BCR-ABL, in accordance with the immunophenotype and molecular biological characteristics of Ph~ ALL. Conclusion: a new Ph~ ALL mouse model was successfully developed, which can provide a powerful tool for the study of Ph~ ALL.
