TRPV3通道在干酵母致发热大鼠下丘脑的表达及机制

【摘要】:目的探讨TRPV3通道在干酵母致发热大鼠下丘脑组织中的表达变化及其对体温的影响。方法采用20%干酵母混悬液背部皮下注射复制发热大鼠模型(模型组),对照组大鼠皮下注射相应体积的生理盐水。观察大鼠的体温、下丘脑组织Ca~(2+)浓度和环磷酸腺苷的变化;通过RT-PCR和Western blot法检测TRPV3通道在下丘脑中的表达变化趋势,分析其与体温之间的相关性。结果模型组大鼠的体温在注射干酵母混悬液后明显上升(P0.01)。模型组大鼠下丘脑组织环磷酸腺苷与对照组比较显著增加(P0.01),Ca~(2+)浓度差异无统计学意义(P0.05)。模型组大鼠下丘脑组织TRPV3表达趋势与对照组比较显著增多(P0.05);TRPV3蛋白表达量变化与体温变化值呈正相关(P0.05)。结论干酵母导致大鼠发热过程中,下丘脑组织TRPV3通道被激活,参与体温调节。
[Abstract]:Objective to investigate the expression of TRPV3 channel in hypothalamus of rats with fever induced by dry yeast and its effect on body temperature. Methods Fever rats were subcutaneously injected with 20% dry yeast suspension (model group), and rats in control group were subcutaneously injected with normal saline. The changes of temperature, the concentration of Ca~ (2) and cyclic adenosine monophosphate in hypothalamus of rats were observed, and the expression trend of TRPV3 channel in hypothalamus was detected by RT-PCR and Western blot methods, and the correlation between TRPV3 channel and body temperature was analyzed. Results the body temperature of the model group increased significantly after injection of dry yeast suspension (P0.01). The concentration of cyclic adenosine monophosphate in the model group was significantly higher than that in the control group (P0.01). There was no significant difference in the concentration of Ca~ (2) between the model group and the control group (P0.05). The expression trend of TRPV3 in hypothalamus of the model group was significantly higher than that of the control group (P0.05), and the expression of TRPV3 protein was positively correlated with the change of body temperature (P0.05). Conclusion in the process of fever induced by dry yeast, the TRPV3 channel in hypothalamus is activated and participates in the regulation of temperature.
