伤寒沙门菌SNPs分子分型分析和鼠伤寒沙门菌分子流行病学特点研究

【摘要】:伤寒沙门菌是引起伤寒的病原菌,目前全球尤其在发展中国家仍然是严重的公共卫生问题。我国把伤寒列为乙类传染病,是重点防控的传染病。通过分子分型的方法能够了解我国和国际伤寒沙门菌的分子流行病学特点,为传染病的监测、暴发识别和追踪传染源提供背景信息。本研究分别应用26个基因位点的SNPs分型和PulseNet推荐的沙门菌PFGE分型方案对我国1959—2007年的伤寒沙门菌进行分型,同时比较国内外沙门菌的SNPs分型特点。研究结果表明选取的SNPs基因位点适合我国伤寒沙门菌株的分型,我国的伤寒沙门菌具有遗传多态性。 鼠伤寒沙门菌是一种常见的食源性病原菌,可引起人类胃肠炎等疾病。了解近几年我国鼠伤寒沙门菌临床分离株的分子分型特点,为以实验室为基础的食源性疾病暴发的污染源及传播途经的追踪提供基线数据。根据国际PulseNet公布的沙门菌PFGE分型方案及7个位点的鼠伤寒沙门菌MLVA分子分型方法,对2006-2010年分离自我国七个省(直辖市)的294株鼠伤寒沙门菌进行两种方法的分子分型分析。这些菌株先经XbaⅠ酶切,脉冲场凝胶电泳后,获得87种带型,其分辨率(D值)为0.9050。对其中的主要优势带型JPXX01.CN0001菌株76株、JPXX01.CN0006菌株40株和JPXX01.CN0073菌株24株进一步用第二种限制性内切酶BlnⅠ酶切后,分别获得10、22和17种带型。采取MLVA分析,获得198种带型, D值是0.9929。对流行病学调查显示为暴发病人和食物来源的菌株进行PFGE双酶切及MLVA分析,两种实验分析结果获得一致性结果,,均显示这些菌株具有明显的聚集性。两种分子分型结果显示我国鼠伤寒沙门菌临床分离株具有遗传多样性,MLVA分型方法的分辨能力高于PFGE,在确认鼠伤寒沙门菌引起的暴发事件时,采用需时较短,操作方便的MLVA分型方法可满足菌株聚集性分析。
[Abstract]:Salmonella typhimurium is the pathogen of typhoid fever, and it is still a serious public health problem all over the world, especially in developing countries. Our country classified typhoid fever as a class B infectious disease, is the key prevention and control infectious disease. The molecular epidemiological characteristics of Salmonella typhimurium in China and the world can be understood by molecular typing method, which can provide background information for the surveillance of infectious diseases, the identification of outbreaks and the tracing of the source of infection. In this study, SNPs typing of 26 loci and PFGE typing of Salmonella typhimurium recommended by PulseNet were used to classify Salmonella typhimurium from 1959 to 2007 in China. At the same time, the characteristics of SNPs typing of Salmonella typhimurium at home and abroad were compared. The results showed that the selected SNPs gene locus was suitable for the typing of Salmonella typhimurium in China, and Salmonella typhimurium had genetic polymorphism in China.
