胶原液晶膜对细胞生长的影响及其仿生矿化研究

【摘要】:液晶是一种介于液相和固相的中间相,,是自然界流动性与有序性的有机结合,液晶态普遍存在于生物体内。骨组织中的大多数矿物质晶体的形成都和胶原纤维密切相关。系统研究液晶胶原的形成与功能及其与无机钙磷盐的有机复合,对开发新型仿生材料和人工组织器官研究有重要意义。 本研究从仿生角度出发,制备出胶原液晶膜。采用偏光显微镜、扫描电镜和原子力显微镜研究胶原膜的液晶织构和拓扑结构,通过MTT法、电镜观察、细胞骨架荧光染色观察研究胶原液晶膜对小鼠成纤维细胞3T3及大鼠成骨细胞的黏附、增殖、生长形貌及细胞骨架组装的影响。通过细胞周期分析、碱性磷酸酶及骨钙素测定进一步研究胶原液晶膜对成骨细胞生长及功能表达状况的影响。另一方面研究胶原液晶膜在模拟体液中的矿化情况,探讨钙磷无机矿物在液晶态胶原基质上沉积的过程和机理。结果表明,胶原液晶膜呈胆甾型液晶织构,具有典型螺旋周期结构的胶原纤维按一定方向自组装,排列具有明显取向性;与非液晶膜相比,胶原液晶膜不仅能更好的促进3T3及成骨细胞的黏附、增殖及功能表达,且其拓扑结构对细胞生长具有接触引导效应;胶原液晶膜独特的胶原纤维结构和排列影响无机钙磷晶体的沉积和形成。
[Abstract]:Liquid crystal is a kind of intermediate phase between liquid phase and solid phase. Liquid crystal is an organic combination of fluidity and ordering in nature. Liquid crystal exists in organisms generally. The formation of most mineral crystals in bone tissue is closely related to collagen fibers. A systematic study on the formation and function of liquid crystal collagen and its organic compound with inorganic calcium and phosphorus salts is of great significance for the development of new biomimetic materials and the study of artificial tissues and organs. In this study, collagen liquid crystal membrane was prepared from bionic point of view. The texture and topological structure of collagen films were studied by polarized light microscope, scanning electron microscope and atomic force microscope. The effects of collagen liquid crystal membrane on adhesion, proliferation, growth morphology and cytoskeleton assembly of mouse fibroblast 3T3 and rat osteoblast were observed by cytoskeleton fluorescence staining. Cell cycle analysis, alkaline phosphatase (ALP) and osteocalcin (osteocalcin) were used to study the effects of collagen liquid crystalline membrane on the growth and functional expression of osteoblasts. On the other hand, the mineralization of collagen liquid crystal membrane in simulated body fluid was studied, and the process and mechanism of calcium and phosphorus inorganic mineral deposition on liquid crystalline collagen matrix were discussed. The results show that the collagen liquid crystal membrane is cholesteric liquid crystal texture, and the collagen fibers with typical spiral periodic structure are self-assembled in a certain direction and have obvious orientation. Compared with the non-liquid crystal membrane, the collagen liquid crystal membrane can not only promote the adhesion, proliferation and functional expression of 3T3 and osteoblasts, but also have contact-guiding effect on cell growth. The unique structure and arrangement of collagen fibers affect the deposition and formation of inorganic calcium and phosphorus crystals.
