体外培养对小鼠生精细胞印迹基因H19和IGF2r甲基化状态的影响

【摘要】:目的:通过改进培养条件建立一个稳定、高效的小鼠生精细胞体外培养方法,获得更多的单倍体圆形和长形精子细胞。方法:采用两步酶消化法和差速贴壁法相结合的方法分离生精细胞,以曲细精管来源细胞为饲养层(MSF),在添加睾酮(T)、卵泡刺激素(FSH)、低浓度(10ng/ml)胶质细胞神经营养因子(GDNF)和表皮生长因子(EGF)的DMEM/ F-12培养基中培养生精细胞;采用RT-PCR检测培养前、生精细胞集落形成、出现圆形精子细胞时精母细胞特异性基因P19和单倍体精子细胞特异性基因TP1 mRNA的表达,判断生精细胞增殖和分化情况;同时采用激光共聚焦显微镜(LSCM)进行分选细胞纯度检测。结果:以MSF为饲养层,在添加T、FSH、低浓度GDNF和EGF的情况下,培养第7 d可见较多的生精细胞克隆并可检测到P19 mRNA表达,第12 d可见较多的圆形精子细胞和个别活动精子以及TP1 mRNA的表达;经LSCM鉴定,分选的单倍体圆形精子细胞纯度为82%。结论:本实验所建立的培养体系是一个高效、稳定的生精细胞体外培养体系;激光共聚焦显微镜可用于初步鉴定单倍体精子细胞。 目的:通过对体外培养获取的小鼠单倍体精子细胞印迹基因H19 ICR(Imprinting Control Region)和IGF2r DMR2(Differentially Methylated Region 2)区甲基化状态的检测,初步评估体外培养对生精细胞印迹基因甲基化状态的影响。方法:采用亚硫酸氢盐测序(BSP)技术,以印迹基因H19 ICR和IGF2r DMR2区为扩增的目的区域,对经体外培养获得的单倍体精子细胞进行甲基化修饰后巢式和半巢式PCR扩增,采用DNAman软件对特异性扩增产物进行测序分析,与Gene Bank上相对应的序列进行比对,判断其甲基化状态,并与小鼠附睾内成熟精子进行比较。结果:小鼠成熟精子中H19 ICR区15个CpG位点呈高度甲基化状态(96.67%),IGF2r DMR2区呈非甲基化状态(94.29%);而经体外培养获得的单倍体精子细胞H19 ICR区甲基化程度显著降低(69.33%,P0.01),IGF2r DMR2区甲基化程度显著增加(44.29%,P0.01)。结论:昆明白小鼠成熟精子中H19 ICR区呈高度甲基化状态;IGF2r DMR2区呈非甲基化状态;体外培养产生的单倍体圆形和长形精子细胞中,H19 ICR区发生了广泛的甲基化丢失,IGF2r DMR2区发生了异常甲基化。
