开结汤对腺嘌呤所致慢性肾衰竭模型大鼠TGF-β1、MCP-1表达的影响

【摘要】:目的研究开结汤对腺嘌呤所致慢性肾衰竭模型大鼠TGF-β1、MCP-1表达的影响。方法选取60只SD大鼠按体质量随机分为6组:空白对照组、模型组、开结汤低、中、高剂量治疗组、尿毒清治疗组,除空白组外,其余5组连续灌服腺嘌呤21 d以复制慢性肾衰竭大鼠模型,剂量均是300 mg/(kg·d)。造模成功后用开结汤、尿毒清进行干预,治疗30 d后,心脏取血检测常规生化指标,取大鼠肾组织,用免疫组化方法检测TGF-β1、MCP-1的表达。结果与正常组比较,模型组大鼠血清中BUN、Scr、P的含量及肾组织中TGF-β1、MCP-1的表达显著增高,差异有统计学意义(P0.05);经过药物治疗,开结汤3个剂量组及尿毒清组大鼠血清中BUN、Scr、P的含量及肾组织中TGF-β1、MCP-1的表达明显低于模型组(P0.05);开结汤3个剂量组与尿毒清组比较,低、中剂量组在降低血清BUN、Scr、P的含量及抑制TGF-β1、MCP-1在肾组织中表达方面较尿毒清组显著(P0.05)。结论开结汤可以改善肾功能,保护肾脏。作用途径是抑制TGF-β1及MCP-1在肾组织中的表达,减少炎性细胞浸润,从而延缓肾纤维化进程。
[Abstract]:Objective to study the effect of Kaijie decoction on the expression of TGF- 尾 1 and MCP-1 in rats with chronic renal failure induced by adenine. Methods 60 SD rats were randomly divided into 6 groups according to body mass: blank control group, model group, low, middle and high dose Kaijie decoction treatment group, Niaoduqing treatment group, except blank group, The other five groups were given adenine for 21 days to establish the rat model of chronic renal failure at a dose of 300 mg/ (kg 路d). After successful establishment of the model, Kaijie decoction and Niaoduqing were used to intervene. After 30 days of treatment, the routine biochemical indexes were detected by heart blood collection, the kidney tissue of rats was taken, and the expression of TGF- 尾 1 and MCP-1 was detected by immunohistochemical method. Results compared with the normal group, the content of BUN,Scr,P in serum and the expression of TGF- 尾 1 and MCP-1 in renal tissue in the model group were significantly higher than those in the normal group (P 0.05). After drug treatment, the content of BUN,Scr,P in serum and the expression of TGF- 尾 1 and MCP-1 in renal tissue of rats in three dose groups of Kaijie decoction and Niaoduqing group were significantly lower than those in model group (P 0.05). Compared with Niaoduqing group, Kaijie decoction three dose groups were lower than Niaoduqing group in decreasing the content of serum BUN,Scr,P and inhibiting the expression of TGF- 尾 1 and MCP-1 in renal tissue (P 0.05). Conclusion Kaijie decoction can improve renal function and protect kidney. The pathway is to inhibit the expression of TGF- 尾 1 and MCP-1 in renal tissue and reduce the infiltration of inflammatory cells, thus delaying the process of renal fibrosis.
