靶向哺乳动物细胞线粒体的核酸转运

【摘要】:线粒体DNA(mitochondrial DNA,mtDNA)的遗传性突变和缺陷是多种线粒体功能失调相关疾病的根本原因。靶向线粒体递送核酸,可从根本上纠正mtDNA突变、挽救mtDNA损伤、阻断疾病进程。哺乳细胞内线粒体的核酸转运途径与细胞核的基因转染大不相同。该文综述了向哺乳动物细胞线粒体递送DNA和RNA(tRNA、rRNA、mRNA和反义RNA)的有效策略,并对其存在问题和发展趋势做一阐述。
[Abstract]:The genetic mutation and defect of mtDNA (DNA (mitochondrial DNA,mtDNA) are the root causes of many diseases related to mtDNA dysfunction. The delivery of nucleic acid to mitochondria can fundamentally correct mtDNA mutation, save mtDNA damage and block the process of disease. The nucleic acid transport pathway of mitochondria in lactation cells is very different from that of nuclear gene transfer. In this paper, the effective strategies of delivering DNA and RNA (tRNA,rRNA,mRNA and antisense RNA) to mammal cells are reviewed, and their existing problems and development trends are described.
