碘液在芽孢染色中的应用研究

【摘要】:目的探寻一种满意的芽孢染色方法 ,同时该方法能克服染料加温后挥发给实验操作人员带来的身体伤害。方法以碘液为媒染剂,用冷染方法替代传统芽孢加温染色,然后进行细菌涂片、干燥、固定、染色。初染液为石炭酸复红5 min,碘液媒染1 min,95%乙醇脱色5 s,碱性美兰复染1 min。比较石炭酸复红染色后加碘液、石炭酸复红染色不加碘液、石炭酸复红加温染色不加碘液的染色效果。结果石炭酸复红染色后加碘液与石炭酸复红加温染色不加碘液的染色效果相同,石炭酸复红染色不加碘液也能看出芽孢,但不明显。结论用碘液助染可获得满意的芽孢染色结果,且用该法可减少石炭酸挥发给实验操作人员带来的危害。
[Abstract]:Objective to explore a satisfactory method of spore staining, which can overcome the physical injury caused by the volatilization of dye after heating. Methods iodine solution was used as mordant, cold staining method was used instead of traditional spore heating staining, and then bacterial smear, drying, fixation and staining were carried out. The initial dye was decolorized by 1 min,95% ethanol and 1 min. by carbonic acid red 5 min, iodine solution mordant staining for 5 s and alkaline Meilan restaining for 1 min.. The dyeing effects of iodized solution, carbonic acid red staining without iodine solution and carbonic acid rered staining without iodine solution were compared. Results the staining effect of adding iodine solution was the same as that of carbonic acid rered staining without iodine solution. Spores could also be seen by carbonic acid red staining without iodine solution, but it was not obvious. Conclusion satisfactory spore staining results can be obtained with iodine solution, and the harm caused by carbonic acid volatilization to experimental operators can be reduced by this method.
