FasL对树突状细胞功能的调控及在真菌感染中的效应研究

树突状细胞(dendritic cells,DC)是由Steinman和Cohn在小鼠脾脏中所发现的一种具有树枝状突起的独特形态的细胞。DC是目前已知的最强的抗原提呈细胞,它是特异性免疫应答的触发者,因为它能够刺激初始型T细胞(na?ve T cell)活化和增殖,而M?、B细胞等只能触发已活化的T细胞或记忆性T细胞发生免疫应答。而且也有报道表明,活化的DC细胞能产生释放大量的炎症因子TNF-α和IL-6等。而大量炎性因子的释放促进全身炎症反应综合征的发生发展进一步诱导多器官功能衰竭。故对可调控DC功能的相关分子进行研究具有重要意义。同时,细胞凋亡与固有免疫的关系仍是目前研究的热点,凋亡相关分子对固有免疫的调节效应也逐渐被人们所关注。以往研究表明,作为TNFR和TNF家族重要成员的Fas和FasL分子是经典的凋亡诱导信号,但近年来的研究表明,Fas和FasL也可以分别传递信号,发挥不同于凋亡诱导的效应。FasL分子与Fas受体结合后,表达FasL分子的细胞其自身在FasL分子反向信号的作用下发生相应改变。相关的体内和体外实验也证明,FasL分子作为共刺激分子参与胸腺细胞成熟和成熟T细胞...

军医大学硕士学位论文) FasL 突变小鼠来源的 BMDC 对 CLR 受体刺激剂的敏感性降低为了研究 DC 的 FasL 反向信号在抗真菌感染中的效应,我们将来源于野生型小鼠sL 胞内段突变小鼠 FasL△Intra 来源的 BMDC 用 LPS ,低浓度的 zymd(dectin-1 受活剂)和 mannan(dectin-2 受体激活剂)处理后,ELISA 检测细胞因子 IL-6,TNF分泌,结果发现 FasL△Intra 小鼠来源的 BMDC 对 CLR 受体激动剂不敏感,IL-6,TNF分泌明显低于 WT 小鼠来源的 BMDC(图 3),这说明 DC 在低剂量 CLR 受体激动激下,FasL 反向信号可能促进 CLR 信号通路的活化。
