反式脂肪酸通过SIRT1/PGC1-α/Nrf2信号通路介导的大鼠血管损伤

目的:本次研究主要致力于观察摄入低剂量反式脂肪酸是否会对血管造成损伤及其可能的机制。方法:8周龄清洁级SD大鼠48只,雌雄各半。大鼠适应性喂养一周,遵从随机、均衡原则,随机分为3组:(1)对照组(ND);(2)1%TFA饮食组(LTD);(3)4%TFA饮食组(HTD),每组16只,自由进食、进水,雌雄分笼,饲养12周。实验期间每周测量体重及进食量,并定期测量血压。连续喂养12周后,断头处死,收集血液,分离血清用于血脂检测。分离出大鼠的血管组织,其中每组6只用于做病理学切片,观察血管形态学变化,每组取3只用于检测血管组织内ROS生成情况,余下血管组织采用Western blot法测定大鼠血管组织SIRT1、PGC1-α、PPARγ、NOX4、Nrf2、Bcl-2、Bax、caspase3和cleaved-caspase3蛋白的表达量。数据以`x±SD表示,多组间比较采用SPSS 20.0软件对数据进行One-way ANOVA检验,P<0.05为差异有统计学意义。结果:1.实验结束时,各组大鼠体重及进食量均随年龄增加而增加,且各组间无统计学差异(P>0.05)。各组大鼠于0...
