茶黄素纳米粒雾化吸入对大鼠炎性气道黏蛋白5AC的调控

【摘要】:目的:探讨高频雾化吸入茶黄素乳酸-羟基乙酸共聚物(poly lactic-co-glycolic acid,PLGA)纳米粒对炎性气道黏蛋白5AC(mucoprotein 5AC,MUC5AC)的作用。方法:通过改良法制备茶黄素PLGA纳米粒,用粒径分析仪检测纳米粒平均粒径,并进行包封率和载药量测定及体外释放试验。将SD大鼠分为3组:对照组(A组)、香烟吸入组(B组)及干预组(C组)。RT-PCR检测各组黏蛋白5AC m RNA含量;酶联免疫吸附测定法(ELISA)观察MUC5AC蛋白表达的变化,并进行组化对比大鼠气道组织黏蛋白变化情况。结果:制备的茶黄素PLGA纳米粒性质稳定,测得其的平均粒径为(108.24±2.86)nm,适宜雾化吸入。香烟吸入组MUC5AC的蛋白表达明显升高,较对照组有显著性差异(P=0.000),而给予茶黄素PLGA纳米粒处理后,与香烟组比,指标明显下调(P=0.001)。MUC5AC m RNA表达的RT-PCR电泳结果也有类似结果。气道组织化学PAS染色也提示雾化吸入茶黄素PLGA纳米粒后,气道表面黏蛋白表达显著下降。结论:通过改良法制备茶黄素PLGA纳米粒性质稳定,大小均匀;经雾化途径吸入可发挥抑制炎性气道的黏蛋白5AC的高分泌作用,为茶黄素的开发应用提供了新的途径。
[Abstract]:Objective: to study the high frequency atomization inhalation of theaflavin-lactic acid-glycolic acid copolymer poly lactic-co-glycolic acid. The effect of PLGA nanoparticles on inflammatory airway mucoprotein 5ACmucoprotein 5ACMUC5AC. Methods: theaflavin PLGA nanoparticles were prepared by modified method. The average particle size was measured by particle size analyzer, encapsulation efficiency, drug loading and in vitro release test were performed.
