染矽尘大鼠结缔组织生长因子和Ⅱ型肺泡细胞表面抗原的表达

【摘要】:[目的]探讨结缔组织生长因子(CTGF)与Ⅱ型肺泡细胞表面抗原(KL-6)在染矽尘大鼠血清、支气管肺泡灌洗液(BALF)及肺组织中的表达及意义。[方法]将49只Wistar雄性大鼠随机分为对照组和染尘后7、14、21、28、60、90 d共7个组,每组7只,染尘组采用非气管暴露法灌注二氧化硅悬浮液(100 mg/m L),对照组以同样方法灌注生理盐水,取其肺组织并采集血清及BALF。采用免疫组织化学法测定肺组织中CTGF和KL-6的表达水平;采用酶联免疫吸附法(ELISA)检测血清及BALF中CTGF、KL-6的质量浓度。[结果]免疫组化结果显示,染尘大鼠肺组织中CTGF和KL-6呈阳性表达,且染尘组的平均光密度值均高于对照组,差异有统计学意义(P0.05);染尘后随时间的延长,CTGF表达呈先增后降的趋势,在染尘后28 d达到最高峰,差异有统计学意义(P0.05);KL-6呈持续上升趋势,且差异有统计学意义(P0.05)。ELISA结果显示,染尘大鼠血清及BALF中CTGF质量浓度的变化趋势基本与其在肺组织中的一致,但是在染尘后21 d达到峰值;KL-6在血清及BALF中的变化也基本与肺组织中一致。[结论]CTGF和KL-6与肺纤维化有密切的关系,在矽肺形成过程中发挥了作用。
[Abstract]:[Objective] to investigate the relationship between CTGF and KL-6 in serum of rats exposed to silica dust. Expression and significance of BALF in bronchoalveolar lavage fluid and lung tissue. [Methods] Forty-nine male Wistar rats were randomly divided into 7 groups, 7 in each group. In the dust-infected group, the silica suspension was perfused with 100 mg/m / L of silica suspension by non-tracheal exposure, while the control group was perfused with normal saline in the same way. The expression of CTGF and KL-6 in lung tissue was determined by immunohistochemical method. Enzyme linked immunosorbent assay (Elisa) was used to detect the concentration of CTGFKL-6 in serum and BALF. [Results: the positive expression of CTGF and KL-6 was found in the lung tissues of the dust-exposed rats. The average optical density of the dust-exposed rats was higher than that of the control group (P 0.05). The expression of CTGF increased first and then decreased with time, and reached its peak at 28 days after dust exposure, the difference being statistically significant (P 0.05). KL-6 showed an increasing trend, and the difference was statistically significant (P 0.05). Elisa results showed that. The change trend of CTGF concentration in serum and BALF of dust-exposed rats was basically the same as that in lung tissue, but reached the peak at 21 days after dust exposure. The changes of KL-6 in serum and BALF were similar to those in lung tissue. [Conclusion CTGF and KL-6 are closely related to pulmonary fibrosis and play an important role in silicosis formation.
