COPD患者肺泡及外周血单核源性巨噬细胞Cofilin的表达与其吞噬烟曲霉功能的相关性

【摘要】:目的探讨慢性阻塞性肺疾病(COPD)患者肺泡及外周血单核源性巨噬细胞Cofilin的表达与其吞噬烟曲霉功能的相关性。方法选取2015年7月至2016年5月南方医科大学珠江医院呼吸内科门诊20名COPD患者,其中观察组1(2011版GOLD中A和B组),观察组2(2011版GOLD中C和D组),健康对照组10人。3组行支气管肺泡灌洗术、采取外周血,分别提取AM和MDM,检测各组的AM和MDM对烟曲霉孢子的吞噬能力。运用实时荧光定量(qRT-PCR)和免疫蛋白印迹实验(Western blot)检测3组Cofilin蛋白的表达,比较其差异。结果 3组人群MDM/AM吞噬烟曲霉孢子的菌落数分别为(17±3)个、(16±2)个、(42±3)个(F=73.446,P0.001),两观察组与健康组差异明显,而两观察组无明显差异。qRT-PCR和Western blot结果显示,两观察组中Cofilin蛋白表达明显高于健康组。结论 COPD患者肺泡及外周血单核源性巨噬细胞吞噬烟曲霉功能降低可能与Cofilin蛋白升高有关。
[Abstract]:Objective to investigate the relationship between the expression of Cofilin in alveolar and peripheral blood mononuclear macrophages and the phagocytic function of Aspergillus fumigatus in patients with chronic obstructive pulmonary disease (COPD). Methods from July 2015 to May 2016, the Pearl River Hospital of Southern Medical University was selected. Twenty patients with COPD in respiratory medicine outpatient department, Group A and B of GOLD were treated with bronchoalveolar lavage, group C and group D were treated with bronchoalveolar lavage and peripheral blood was treated with peripheral blood. The phagocytic ability of AM and MDM to Aspergillus fumigatus spores was determined by AM and MDM, and the expression of Cofilin protein was detected by real-time fluorescence quantitative analysis and Western blotanalysis. Results the colony number of MDM/AM phagocytic Aspergillus fumigatus spores in the three groups was 17 卤3), 16 卤2) and 42 卤3 respectively. The difference between the two groups was significant, but there was no significant difference between the two groups. QRT-PCR and Western blot showed that there was no significant difference between the two groups. Conclusion the decrease of pulmonary alveolar and peripheral blood mononuclear macrophages phagocytosis of Aspergillus fumigatus in patients with COPD may be related to the increase of Cofilin protein.
