二硫代氨基吡咯烷对大鼠急性肺损伤保护作用

【摘要】:目的探讨二硫代氨基吡咯烷(PDTC)对内毒素(LPS)诱导急性肺损伤(ALI)大鼠肺组织的保护作用及其机制。方法将72只Wistar大鼠随机分为对照组(C组),LPS组(L组),PDTC+LPS组(P组)。各组分为1,3,6,12 h亚组,每组6只。L组和P组制作内毒素性肺损伤模型。P组腹腔注射LPS前30 min尾静脉注射PDTC,120 mg·kg-1,L组和C组给予等体积0.9%氯化钠溶液。观察大鼠肺组织病理变化,肺组织湿/干质量比(W/D)变化,RT-PCR法检测大鼠肺组织细胞间黏附分子-1 mRNA(ICAM-1mRNA)的表达,放射免疫法检测大鼠血清肿瘤坏死因子α(TNF-α)、白细胞介素6(IL-6)含量变化。结果病理切片显示,与C组比较,L组肺组织结构破坏严重,W/D明显增大,ICAM-1mRNA在各时间点表达明显增强(P0.05),血清TNF-α、IL-6含量在1,3,6 h增高(P0.05)。与L组比较,P组肺组织结构破坏明显减轻,W/D减小(P0.05),ICAM-1mRNA表达明显减少(P0.05),血清TNF-α、IL-6含量1,3,6 h降低(P0.05或P0.01)。结论 PDTC能明显抑制ICAM-1mRNA表达,降低TNF-α和IL-6含量,PDTC可能对ALI大鼠有保护作用。
[Abstract]:Objective to investigate the protective effect of dithioaminopyrrolidine on the lung tissue of rats with acute lung injury induced by lipopolysaccharide (LPS) and its mechanism. Methods 72 Wistar rats were randomly divided into control group (group C) and control group (group C). Endotoxin-induced lung injury model was established in each group (n = 6) and group P (n = 6). Group P was injected intraperitoneally with PDT Con 120mg 路kg ~ (-1) L and group C (n = 30) 30 min before intraperitoneal injection with sodium chloride solution of equal volume (0.9% mg 路kg ~ (-1)). The pathological changes of lung tissue in rats were observed. Lung tissue wet / dry weight ratio: reverse transcription-polymerase chain reaction (RT-PCR) was used to detect the expression of intercellular adhesion molecule-1 mRNA (ICAM-1mRNAs), and radioimmunoassay (RIA) was used to detect the contents of tumor necrosis factor- 伪 (TNF- 伪) and interleukin-6 (IL-6) in rat lung tissue. Compared with group C, the expression of ICAM-1 mRNA and the level of TNF- 伪 IL-6 in serum increased significantly at each time point in group C and group C, respectively. Compared with group L, the destruction of lung tissue structure in group C significantly decreased the expression of ICAM-1 mRNA in lung tissue, and the expression of ICAM-1 mRNA decreased significantly in group A (P 0.05) compared with that in group L (P < 0.05), and the expression of ICAM-1 mRNA was significantly reduced in group C than in group C (P < 0.05), and the level of TNF- 伪 -IL-6 in serum was significantly increased at 6 h after treatment. The level of TNF- 伪 IL-6 in serum decreased by P0.05 or P0.01 for 6 h. Conclusion PDTC can significantly inhibit the expression of ICAM-1mRNA. The decrease of TNF- 伪 and IL-6 may have protective effect on ALI rats.
