不同海拔大鼠慢性阻塞性肺疾病模型全身效应与血浆TNF-α、IL-6、IL-10的关系

【摘要】:目的:探讨不同海拔大鼠慢性阻塞性肺疾病(COPD)模型全身效应的差别及与血浆中肿瘤坏死因子α(TNF-α)、白细胞介素6(IL-6)及白细胞介素10(IL-10)的水平的关系,进而了解高原缺氧对COPD大鼠全身炎症反应的影响。 方法:以腹腔注射香烟提取物法建立大鼠COPD模型。50只SD大鼠随机分为对照组、中海拔盐水组、中海拔COPD组(西宁,海拔2260m)和高海拔盐水组、高海拔COPD组(低压氧舱,海拔5500m),,观察大鼠进食、饮水、活动量、情绪及体重,应用双抗体夹心法(ELISA)方法测定SD大鼠血浆中TNF-α、IL-6及IL-10的水平。 结果:不同组别大鼠全身表现不同,中海拔COPD组较中海拔盐水组及对照组活动量减少,易激惹;高海拔盐水组较中海拔盐水组,高海拔COPD组较中海拔COPD组饮食、饮水量及活动量明显减少,且易激惹。高海拔盐水组体重低于中海拔盐水组及对照组(P0.01),高海拔COPD组体重低于中海拔COPD组(P0.01)。高海拔盐水组TNF-α、IL-6水平明显高于中海拔盐水组和对照组(P0.05);高海拔COPD组TNF-α、IL-6水平高于中海拔COPD组(P0.05),同海拔COPD组高于盐水组(P0.05)。高海拔盐水组IL-10高于中海拔盐水组及对照组(P0.01) 结论:1.高原缺氧促进健康大鼠血浆中TNF-α、IL-6的分泌并可造成全身效应;2.高原缺氧加剧了COPD大鼠的全身炎症反应和全身效应;3.IL-10在COPD大鼠中减弱了抑制炎症因子、抑制炎症反应的作用。
[Abstract]:Objective: To investigate the effects of chronic obstructive pulmonary disease in rats at different altitudes (COPD) model and the difference between the systemic effects and plasma tumor necrosis factor alpha (TNF- alpha), interleukin 6 (IL-6) and interleukin 10 (IL-10) the level of the relationship, and to understand the effects of hypoxia on systemic inflammatory reaction COPD rats.
Methods: intraperitoneal injection of cigarette smoke extract method to establish a rat model of COPD.50 SD rats were randomly divided into control group, saline group in altitude, altitude COPD group (Xining, 2260m above sea level) and high altitude saline group, COPD group (high altitude hypobaric chamber, altitude 5500m), rats eating, drinking water that activity, mood and body. The application of double antibody sandwich enzyme-linked immunosorbent assay (ELISA) method for the determination of TNF- alpha SD in rat plasma, IL-6 and IL-10 levels.
Results: all rats in different groups were different in the elevation of COPD group compared with the saline group and control group in the altitude of reduced activity, irritability; high altitude saline group than in saline group at high altitude, COPD group than in COPD group at eating, drinking and activity were significantly decreased, and irritability. High altitude elevation weight lower than the saline group saline group and control group (P0.01), high altitude group COPD COPD group weighed less than in altitude (P0.01). The high altitude control group TNF- alpha, IL-6 level was significantly higher than that in soil saline group and control group (P0.05); high altitude group COPD TNF- alpha, IL-6 level is higher than that in the elevation of COPD group (P0.05), with an elevation of COPD group was higher than that of saline group (P0.05). The high altitude is higher than that of saline group IL-10 elevation saline group and control group (P0.01)
Conclusion: 1.. Hypoxia at high altitude promotes the secretion of TNF- and IL-6 in healthy rats, and can cause systemic effects. 2., hypoxia at high altitude aggravates the systemic inflammatory response and systemic effect in COPD rats. 3.IL-10 weakens the inhibition of inflammatory factors and inhibits inflammatory response in COPD rats.
