骨髓间充质干细胞培养液对哮喘小鼠肺部炎症反应的影响

【摘要】:目的: 探索通过向腹腔内注射骨髓间充质干细胞(BMSCs)培养液是否能够改善支气管哮喘小鼠肺部炎症的病理反应的程度,能否下调肺泡灌洗液及血清中T淋巴细胞相关的细胞因子的水平。 方法:随机将30只实验小鼠分为三组,,每组10只: 1.对照组:实验用药物均用注射用生理盐水代替(腹腔内注射及雾化吸入药物)。 2.哮喘组;制作哮喘小鼠动物模型,不予以干预。 3.治疗组;在每次激发前的2小时将2ml的骨髓间充质干细胞培养液向小鼠的腹腔内注射。这样一共激发三次,最后一次激发后48小时将实验小鼠处死,采用病理方法分析收集到的小鼠的血清、肺泡灌洗液以及肺脏组织。 结果: 1.哮喘组小鼠的肺组织与正常的对照组相比,出现明显的变态反应性炎症反应(Underwood评分显著升高,P0.01) 2.治疗组的小鼠与哮喘组相比,肺部变态反应性炎症反应显著减轻(Underwood评分降低,P0.01) 3.哮喘组与对照组相比,小鼠肺泡灌洗液中细胞总数显著增多(P0.01)。 4.治疗组的小鼠与正常对照组相比,肺泡灌洗液中的细胞总数增多(P0.05);而与哮喘组的小鼠相比,肺泡灌洗液中的细胞总数减少(P0.05)。 5.哮喘组小鼠与对照组相比,血清及肺泡灌洗液中IL-17水平显著升高(P0.01);BMSCs培养液治疗组小鼠血清及肺泡灌洗液中IL-17水平比哮喘组显著降低(P0.05)。 6. BABL/c小鼠的血清以及肺泡灌洗液中的IL-4水平,在正常对照组、哮喘组以及治疗组中无统计学差别。 结论: 将BMSCs培养液注于哮喘小鼠腹腔内,可以减轻其肺部炎性病理反应的程度,下调肺泡灌洗液及血清中T淋巴细胞相关的IL-17的水平。
[Abstract]:Objective:To explore whether intraperitoneal injection of bone marrow mesenchymal stem cells (BMSCs) can improve the degree of pulmonary inflammation in asthmatic mice and down-regulate the levels of cytokines associated with T lymphocytes in alveolar lavage fluid and serum.Methods: thirty experimental mice were randomly divided into three groups with 10 mice in each group.1.Control group: all the experimental drugs were replaced by normal saline (intraperitoneal injection and atomization inhalation).2.Asthma group; make asthma mice model, no intervention.3.In the treatment group, bone marrow mesenchymal stem cells (BMSCs) of 2ml were injected intraperitoneally into mice 2 hours before each challenge.The mice were killed 48 hours after the last stimulation. The collected serum, alveolar lavage fluid and lung tissue were analyzed by pathological method.Results:1.Compared with the normal control group, the lung tissue of the asthmatic mice showed obvious allergic inflammatory response, the Underwood score increased significantly (P 0.01).2.Compared with asthma group, lung allergic inflammation was significantly reduced in treatment group (P 0.01).3.Compared with the control group, the total number of cells in the alveolar lavage fluid increased significantly in the asthma group (P 0.01).4.Compared with the control group, the total number of cells in the alveolar lavage fluid increased in the treatment group (P 0.05) and decreased in the lung alveolar lavage fluid (P 0.05) compared with that in the asthma group.5.Compared with the control group, the level of IL-17 in serum and alveolar lavage fluid in asthma group was significantly higher than that in asthmatic group. The level of IL-17 in serum and alveolar lavage fluid in treatment group was significantly lower than that in asthma group.6.The levels of IL-4 in serum and alveolar lavage fluid of BABL/c mice were not significantly different in normal control group, asthma group and treatment group.Conclusion:Injecting BMSCs culture medium into the abdominal cavity of asthmatic mice could reduce the degree of pulmonary inflammatory pathological reaction and down-regulate the level of T lymphocyte related IL-17 in alveolar lavage fluid and serum.
