知母皂苷对肥大细胞活化影响及机制研究

【摘要】:目的: 本研究利用哮喘模型小鼠,观察知母皂苷(Saponins from Anemarrhena Asphodeloides Bunge, SAAB)对Thl和Th2细胞因子抑制分泌的影响。SAAB具有明显的抗炎、抗氧化等药理作用。尽管已证明知母皂苷具有很强的抗炎效果,但是对过敏性哮喘的治疗作用及其机制尚未见报道。本实验将观察知母皂苷对对肥大细胞脱颗粒的影响,探讨知母皂苷抗哮喘作用及其可能的作用机制。 方法: 通过Anti-DNP IgE诱导大鼠被动皮肤过敏反应(PCA)实验,利用伊文思蓝染料观察血管通透性,利用Anti-DNP IgE诱导肥大细胞脱颗粒,体外获取大鼠腹腔肥大细胞,并分成五组,正常组,脱颗粒组,SAAB低浓度组(SAAB25); SAAB中浓度组(SAAB50); SAAB高浓度组组(SAAB100),用比色法测定SAAB在体内对肥大细胞影响,体外实验观察SAAB对Anti-DNP IgE诱导组胺释放、细胞内钙摄入及炎性介质TNF-的影响。 结果: SAAB25~100μg/L浓度对肥大细胞吸光度无明显影响,表明SAAB对肥大细胞的存活几乎无影响。Anti-DNP IgE能明显诱导肥大细胞脱颗粒,给予25μg/LSAAB预处理也可见明显的肥大细胞脱颗粒,但50和100μg/L预处理肥大细胞脱颗粒不明显,能明显的抑制肥大细胞脱颗粒。正常组肥大细胞释放组胺较少。与正常组比较,肥大细胞释放组胺明显增多(P0.05),与脱颗粒组比较SAAB各治疗组均能抑制肥大细胞的组胺释放,其中高、中(100、50μg/L)剂量组与模型组比较,有统计学意义(P0.05)。正常组伊文思蓝渗出较少。正常组伊文思蓝渗出很少。与正常组比较,模型组伊文思蓝渗出明显增多(P0.05),与模型组比较SAAB各治疗组均能减少染料的渗出,其中高、中(300、150mg/kg)剂量组与模型组比较,有统计学意义(P0.05)。正常组肥大细胞钙摄入量较少。与正常组比较,脱颗粒组肥大细胞钙摄入量明显增多(P0.05),与脱颗粒组比较SAAB提取物各治疗组均能抑制肥大细胞钙摄入量,其中高、中(100、50μg/L)剂量组与模型组比较,有统计学意义(P0.05)。 结论: (1)SAAB明显抑制肥大细胞脱颗粒和组胺的释放。 (2)SAAB明细抑制肥大细胞介导的大鼠血浆漏出。 (3)SAAB通过影响钙离子内流抑制肥大细胞脱颗粒。
[Abstract]:Aim: to investigate the effects of saponin Saponins from Anemarrhena Asphodeloides Bunge.Saab on the inhibition of Thl and Th2 cytokines in asthmatic mice. Although it has been proved that Anemarrhenoside has a strong anti-inflammatory effect, the therapeutic effect and its mechanism of allergic asthma have not been reported. In this experiment, we observed the effect of Anemarrhenoside on mast cell degranulation, and discussed the antiasthmatic effect of Aemarrhenoside and its possible mechanism. Methods: Anti-DNP IgE induced passive skin allergic reaction (PCA) in rats. The vascular permeability was observed by Evans blue dye, mast cell degranulation was induced by Anti-DNP IgE, and rat peritoneal mast cells were obtained in vitro and divided into five groups. Normal group, degranulated group, SAAB low concentration group, SAAB medium concentration group, SAAB50, SAAB high concentration group, SAAB100, the effect of Saab on mast cells in vivo was determined by colorimetry, and the effect of Saab on anti-DNP IgE induced histamine release was observed in vitro. Effects of intracellular calcium intake and inflammatory mediator TNF-. Results: SAAB 25 渭 g / L concentration had no significant effect on mast cell absorbance, suggesting that Saab had little effect on mast cell survival. Anti-DNP IgE could induce mast cell degranulation. Pretreatment with 25 渭 g / L SAAB also showed obvious mast cell degranulation, but 50 渭 g / L and 100 渭 g / L pretreatment was not obvious, and could inhibit mast cell degranulation obviously. In normal group, mast cells released less histamine. Compared with the normal group, mast cell released histamine increased significantly (P 0.05). Compared with the degranulation group, Saab could inhibit the histamine release of mast cell. The histamine release of mast cell was inhibited by SAAB. The histamine release of mast cell was significantly higher in the treatment group than that in the model group (P 0.05), and the dose of 50 渭 g / L was higher than that in the model group. In normal group, Evans blue exudate less. In the normal group, Evans blue exudates little. Compared with the normal group, the exudation of Evans blue in the model group was significantly higher than that in the model group. Compared with the model group, Saab treatment group could reduce the dye exudation, especially in the high dose group (300 mg / kg) compared with the model group (P 0.05). The calcium intake of mast cells in normal group was less. Compared with the normal group, the calcium intake of mast cells in the degranulated group was significantly increased (P 0.05). Compared with the degranulated group, Saab extract could inhibit the calcium intake of mast cells in all the treatment groups. The calcium intake of mast cells in the treatment group was higher than that in the model group. Conclusion: Saab can inhibit mast cell degranulation and histamine release. It can inhibit mast cell induced plasma leakage in rats. It can inhibit mast cell degranulation through the influence of calcium influx.
