油酸致急性肺损伤乳猪模型的建立

【摘要】:目的: 采用油酸溶于等容无水酒精混合猪血清持续匀速注射方法,构建符合肺损伤标准、血流动力学稳定的乳猪急性肺损伤动物模型。 方法: 采用改良法构建油酸型乳猪急性肺损伤模型:选用高纯度油酸,以每公斤体重0.1ml的剂量溶于等容无水酒精,然后与10ml自制猪血清经涡旋振荡器充分混匀后,再用微泵注射器经乳猪颈静脉插管在15min内缓慢匀速的注入。出生25-30天的乳猪12只,随机分为两组:a)实验组(Study group,n=6):油酸组,用上述方法注射油酸。b)对照组(Control group,n=6):生理盐水组。用上述方法注射等量生理盐水。注射过程中检测Pa02,当Pa02/FiO2比值300mmHg,即认为模型建立成功。监测肺功能指标如动脉血氧分压、动脉血二氧化碳分压,PH值及血流动力学指标如心率、平均动脉血压、心输出量、中心静脉压;实验结束,取右下肺组织标本,生理盐水冲洗标本,甲醛固定标本,放于容器中,行病理检查。了解其组织学改变情况。 结果: 对照组肺功能指标动脉血氧分压、动脉血二氧化碳分压、pH值及血流动力学指标心率、平均动脉压、心输出量、中心静脉压基础值无显著性差异(P0.05)。持续匀速注射油酸完成后,实验组很快即达到急性肺损伤模型要求。Pa02/Fi02比值下降明显,平均动脉血压、心率、心输出量、动脉氧分压、pH值与对照组相比明显降低(P0.05);实验组动脉二氧化碳分压、心率比对照组明显升高(P0.05)。病理组织学切片显示实验组病变较对照组明显严重。 结论:1.通过油酸混合猪血清持续匀速注射方法,可以构建血流动力学稳定的乳猪肺损伤模型; 2.通过油酸混合猪血清持续匀速注射方法,能够反映临床典型的急性肺损伤/急性呼吸窘迫综合症(ALI/ARDS)病理表现;以这种模型为基础,可以深入开展ALI/ARDS相关课题的研究。
[Abstract]:Aim: to establish an animal model of acute lung injury in suckling pigs with stable hemodynamics according to lung injury standard by continuous uniform injection of oleic acid in serum of isovolumetric anhydrous alcohol mixed pigs. Methods: the acute lung injury model of oleic acid type suckling pig was established by modified method. High purity oleic acid was selected and dissolved in isovolumetric anhydrous alcohol at the dose of 0.1ml per kilogram of body weight, and then mixed with 10ml pig serum by vortex oscillator. Then micropump syringe was used to infuse the jugular vein of the suckling pig slowly and uniformly into 15min. Twelve suckling pigs born 25-30 days were randomly divided into two groups: control group (n = 6): study group (n = 6): oleic acid group (n = 6): control group (n = 6): normal saline group (n = 6). The same amount of saline was injected with the above method. Pa02 was detected during injection, and when the ratio of Pa02 / FiO2 was 300 mmHg, the model was considered successful. Pulmonary function parameters such as arterial partial pressure of oxygen, arterial blood pressure of carbon dioxide and PH value and hemodynamic indexes such as heart rate, mean arterial blood pressure, cardiac output, central venous pressure were monitored. The specimens were washed with normal saline, fixed by formaldehyde and placed in containers for pathological examination. Learn about the histological changes. Results: in the control group, there was no significant difference in arterial oxygen pressure, arterial carbon dioxide pressure and pH value, heart rate, mean arterial pressure, cardiac output and central venous pressure (P0.05). After continuous and uniform injection of oleic acid, the experimental group reached the requirement of acute lung injury model quickly. The ratio of Pa02 / Fi02 decreased significantly, the mean arterial blood pressure, heart rate, cardiac output, arterial oxygen pressure and pH value decreased significantly compared with the control group (P 0.05). The arterial carbon dioxide partial pressure and heart rate in the experimental group were significantly higher than those in the control group (P 0.05). Histopathological sections showed that the pathological changes in the experimental group were significantly more serious than those in the control group. Conclusion 1. The hemodynamic stable lung injury model of suckling pig can be established by continuous uniform injection of oleic acid mixed pig serum. 2. Continuous uniform injection of oleic acid mixed pig serum can reflect the typical clinical manifestations of acute lung injury / acute respiratory distress syndrome (Ali / ARDS), and based on this model, the research on ALI / ARDS related research can be carried out in depth.
