香烟烟雾暴露对大鼠肺组织血小板衍生生长因子-BB及核因子-κB表达的影响

【摘要】:目的:研究烟雾暴露对大鼠肺组织血小板衍生生长因子-BB (Platelet derived growth factor-BB, PDGF-BB)和核因子-kB(nuclear factor-KB, NF-kB)p65表达的影响,探讨烟雾暴露致大鼠肺动脉高压的发生机制。 方法:雄性Wistar大鼠32只,随机分为对照组、烟雾暴露4、8、12周组,每组8只,制备烟雾暴露组肺动脉高压模型。将烟雾暴露组大鼠置于自制实验性大鼠被动吸烟箱内进行被动吸烟,分别烟雾暴露4、8、12周,对照组除无烟雾暴露外,其他条件完全相同。各组大鼠模型制备成功后,右心导管法测大鼠肺动脉平均压(mean pulmonary arterial pressure, mPAP),无菌条件下完整游离心、肺组织,计算大鼠右心肥厚指数(right ventricle hypertrophy index, RVHI),取左肺组织置于-80℃冰箱冻存,用RT-PCR测各组大鼠肺组织mRNA的相对表达量,取右肺组织常规石蜡包埋、切片,用于HE染色,免疫组织化学染色法测各组大鼠肺组织中PDGF-BB及NF-KBp65蛋白的表达,分析各指标的相关性。 结果:①大鼠肺组织中PDGF-BB mRNA相对表达量在烟雾暴露12周组(6.098±0.439)及8周组(4.015±0.052)高于4周组(2.823±0.453)及对照组(1.024±0.231),差异有统计学意义(P均0.05);烟雾暴露12周组(6.098±0.439)高于8周组(4.015±0.052),差异有统计学意义(P0.05);烟雾暴露4周组与对照组组间差异无统计学意义(P0.05);②大鼠肺组织中PDGF-BB蛋白相对表达量在烟雾暴露4周组(0.220±0.005)、8周组(0.367±0.004)、12周组(0.492±0.003)高于对照组(0.077±0.003),差异有统计学意义(P均0.05);烟雾暴露12周组(0.492±0.003)及8周组(0.367±0.004)明显高于4周组(0.220±0.005),差异有统计学意义(P均0.05);烟雾暴露12周组(0.492±0.003)高于8周组(0.367±0.004),差异有统计学意义(P0.05);NF-KBp65蛋白相对表达量在烟雾暴露4周组(0.330±0.003)、8周组(0.452±0.003)、12周组(0.596±0.003)高于对照组(0.186±0.001),差异有统计学意义(P均0.05);烟雾暴露12周组(0.596±0.003)及8周组(0.452±0.003)高于4周组(0.330±0.003),差异有统计学意义(P均0.05);烟雾暴露12周组(0.596±0.003)高于8周组(0.452±0.003),差异有统计学意义(P0.05);③DGF-BB蛋白的表达与PDGF-BB mRNA的相对表达量及与NF-KBp65蛋白的表达均呈正相关(r=0.686、r=0.501,P值均0.01)。 结论:香烟烟雾暴露8周使大鼠肺动脉压力增高,形成肺动脉高压;香烟烟雾暴露4周大鼠肺组织中PDGF-BB表达增高,促使肺血管重塑,至12周渐增高,促使肺动脉高压形成;香烟烟雾暴露4至12周大鼠肺组织中NF-KBp65表达渐增高。烟雾暴露大鼠肺组织中PDGF-BB在转录和翻译水平表达均增高,且PDGF-BB和NF-kB蛋白表达有相关性,提示PDGF-BB和NF-kB在烟雾暴露引起肺动脉平均压增高的过程中可能发挥了一定作用。
[Abstract]:Aim: to investigate the effects of smoke exposure on the expression of platelet derived growth factor-BB (PDGF-BB) and nuclear factor-kB (NF-kB) p65 in rat lung tissue, and to explore the mechanism of pulmonary hypertension induced by smoke exposure. Methods: Thirty-two male Wistar rats were randomly divided into control group (n = 8) and smoke exposure group (n = 8) for 12 weeks. Pulmonary hypertension model was established in smoke exposure group (n = 8). The rats in the smoke exposure group were put into the self-made experimental rats' passive smoking box for passive smoking. The smoke exposure group was exposed to smoke for 4 weeks for 12 weeks. The other conditions were the same in the control group except for no smoke exposure. After the rat model was established successfully, the mean pulmonary artery pressure (mean pulmonary arterial pressure, mPAP) was measured by right cardiac catheterization method. The intact free heart and lung tissues were obtained under aseptic condition. The right heart hypertrophy index (right ventricle hypertrophy index, RVHI) was calculated, and the left lung tissue was frozen in refrigerator at -80 鈩,
