小窝蛋白-1在大潮气量机械通气大鼠肺组织中的表达

【摘要】:目的通过观察不同时间段机械通气大鼠肺组织中小窝蛋白-1(caveolin-1, cav-1)的表达水平,探讨cav-1在呼吸机所致肺损伤(VILI)发病中的作用。 方法雄性SD大鼠32只,被随机分为对照组、大潮气量机械通气半小时组(H-VT0.5h)、大潮气量机械通气1小时组(H-VTlh)和大潮气量机械通气2小时组(H-VT2h)。于光镜下观察各组大鼠肺组织的病理学改变,测定其肺湿/干重比值(W/D)及BALF中总蛋白(TP)含量。各组大鼠肺组织cav-1的表达水平采用免疫组织化学法(SABC法)测定。 结果 1.肺组织cav-1表达结果显示:H-VT0.5h组(0.456±0.055)和对照组(0.394±0.057)均明显高于H-VT2h组(0.297±0.046)和H-VTlh组(0.318±0.051)(P值均0.001),H-VT0.5h组明显高于对照组(P0.001),H-VTlh组明显高于H-VT2h组(P0.009)。 2.肺组织W/D比值结果显示:H-VT2h组(7.172±0.200)和H-VTlh组(5.598±0.227)均明显高于对照组(4.139±0.263)和H-VT0.5h组(4.273±0.197)(P值均0.001),H-VT2h组明显高于H-VTlh组(P0.001),H-VT0.5h组与对照组比较无统计学意义(P0.05)。 3. BALF总蛋白含量测定结果显示:H-VT2h组(0.654±0.0444)和H-VTlh组(0.440±0.0434)均明显高于对照组(0.233±0.0274)和H-VT0.5h组(0.243±0.0416)(P值均0.001),H-VT2h组明显高于H-VTlh组(P0.001),H-VT0.5h组与对照组比较无统计学意义(P0.05)。 4.肺组织HE病理学结果显示:随着机械通气时间延长(尤以1小时后明显),大鼠肺组织损伤程度呈逐渐加重趋势。 结论 1.肺组织cav-1在大潮气量机械通气不同时间段表达水平明显不同,其含量表现为先增高后降低。提示肺组织cav-1高表达对VILI起一定保护作用。 2.大潮气量机械通气可通过抑制肺组织cav-1表达,诱发和加重肺组织损伤。
[Abstract]:Objective to investigate the role of Cav-1 in the pathogenesis of lung injury (VILI) induced by ventilator by observing the expression level of caveolin-1 (Cav-1) in the lung tissue of rats with mechanical ventilation at different time periods.
Methods 32 male SD rats were randomly divided into control group, high tidal volume mechanical ventilation group (H-VT0.5h), large tidal volume mechanical ventilation (H-VTlh) and large tidal volume mechanical ventilation (H-VT2h). The pathological changes of lung tissue in each group were observed under light microscope, and the ratio of lung wet / dry weight (W/D) and total protein in BALF (W/D) and BALF were measured. Content of TP). The expression level of Cav-1 in lung tissue of each group was determined by immunohistochemical method (SABC).
Result
1. Cav-1 expression in lung tissue showed that group H-VT0.5h (0.456 + 0.055) and control group (0.394 + 0.057) were significantly higher than group H-VT2h (0.297 + 0.046) and H-VTlh (0.318 + 0.051) (P value 0.001), and H-VT0.5h group was significantly higher than that of control group (P0.001), and H-VTlh group was significantly higher than group H-VT2h (P0.009).
2. W/D ratio of lung tissue showed that group H-VT2h (7.172 + 0.200) and H-VTlh group (5.598 + 0.227) were significantly higher than that of control group (4.139 + 0.263) and H-VT0.5h group (4.273 + 0.197) (P value 0.001), H-VT2h group was significantly higher than that of H-VTlh group (P0.001), and there was no significant difference between the H-VT0.5h group and the control group (P0.05).
The total protein content of 3. BALF showed that group H-VT2h (0.654 + 0.0444) and H-VTlh group (0.440 + 0.0434) were significantly higher than that of control group (0.233 + 0.0274) and H-VT0.5h group (0.243 + 0.0416) (P value was 0.001), and H-VT2h group was significantly higher than that of H-VTlh group (P0.001), and there was no significant difference between the H-VT0.5h group and the control group (P0.05).
4. the pathological findings of lung tissue HE showed that with the prolongation of mechanical ventilation time (especially after 1 hours), the degree of lung tissue injury gradually increased.
conclusion
The expression level of Cav-1 in 1. lung tissues was significantly different in different time periods of large tidal volume mechanical ventilation, and its content was increased first and then decreased. It suggested that the high expression of Cav-1 in lung tissue has a certain protective effect on VILI.
2. high tidal volume ventilation can induce and aggravate lung injury by inhibiting the expression of Cav-1 in lung tissue.
