支气管哮喘大鼠肺组织中醛糖还原酶表达的变化

【摘要】:目的探讨醛糖还原酶在哮喘大鼠肺组织中表达的变化,以及与气道炎症、气道重塑的关系。 方法健康雄性Wistar大鼠20只,体重210-250克,随机分为两组,即对照组与哮喘组,每组10只。采用卵清白蛋白致敏、激发动物,复制大鼠的哮喘模型。对照组大鼠均以等量生理盐水代替卵清白蛋白。收集肺泡灌洗液,检测白细胞、嗜酸粒细胞计数。肺组织切片HE染色,观察气道形态并检测形态学指标。免疫组织化学法检测醛糖还原酶(Aldose Reductase,AR)在两组大鼠肺组织气道壁的表达水平,采用图像分析系统进行图象分析。 结果哮喘组大鼠肺泡灌洗液中白细胞(WBC)总数及嗜酸性粒细胞(Eos)计数高于对照组(P0.05);哮喘组气道壁面积WA/Pi、气道平滑肌厚度(T1+T2)/Pi×100%较对照组明显增加(P0.05);哮喘组醛糖还原酶的表达水平显著高于对照组(P0.05);哮喘组大鼠肺组织中醛糖还原酶的表达水平与肺泡灌洗液中白细胞总数及嗜酸性粒细胞计数呈正相关(分别为r =0.887, r =0.387,P均0.05 );而且醛糖还原酶的表达水平与WA/Pi、(T1+T2) /Pi×100%呈正相关(分别为r =0.889,r =0.983, P均0.05 )。 结论本实验采用卵清白蛋白(OVA)激发、致敏大鼠成功复制慢性哮喘模型,该模型的主要特征为气道炎症、气道重塑。醛糖还原酶的表达与哮喘的气道炎症、气道重塑呈正相关,,并可能参与气道炎症、气道重塑的过程。
[Abstract]:Objective to investigate the expression of Aldose reductase in lung tissue of asthmatic rats and its relationship with airway inflammation and airway remodeling. Methods Twenty healthy male Wistar rats weighing 210-250g were randomly divided into two groups: control group and asthma group with 10 rats in each group. The asthmatic model of rats was induced by ovalbumin. In the control group, the ovalbumin was replaced by the same amount of normal saline. The alveolar lavage fluid was collected and leukocytes and eosinophils were measured. Lung tissue sections were stained with HE to observe the morphology of the airway and to detect the morphologic indexes. The expression of Aldose reductase (Aldose Reductase,AR) in the airway wall of two groups of rats was detected by immunohistochemical method. Image analysis system was used to analyze the expression of Aldose reductase (Aldose Reductase,AR). Results the total (WBC) count and eosinophil (Eos) count in alveolar lavage fluid of asthmatic rats were higher than those in the control group (P0.05), and the airway smooth muscle thickness (T1T2) / Pi 脳 100% of airway wall area (T1T2) / Pi 脳 100% in asthma group were significantly higher than those in control group (P0.05). The expression of Aldose reductase in asthma group was significantly higher than that in control group (P0.05). The expression of Aldose reductase in lung tissue of asthmatic rats was positively correlated with the total number of leukocytes and eosinophil count in alveolar lavage fluid (r = 0.887, r = 0.387, P 0.05, respectively). The expression level of Aldose reductase was positively correlated with WA/Pi, (T 1T 2) / Pi 脳 100% (r = 0.889r 0.983, P < 0.05). Conclusion the model of chronic asthma was successfully induced by ovalbumin (OVA) in sensitized rats. The main characteristics of the model were airway inflammation and airway remodeling. The expression of Aldose reductase was positively correlated with airway inflammation and airway remodeling, and may be involved in the process of airway inflammation and airway remodeling.
