不同剂量红景天苷对COPD大鼠的骨骼肌功能及其FOXO1表达的影响

【摘要】:目的:探索不同剂量红景天苷对COPD大鼠骨骼肌功能及其FOXO1表达的影响,为红景天苷应用于改善COPD骨骼肌功能障碍提供实验依据。方法:取40只成年SD大鼠分成5组,每组8只,分别为:空白组(C),COPD模型+安慰剂组(CS)、COPD模型+低剂量红景天苷组(CS+L)、COPD模型+中剂量红景天苷组(CS+M)、COPD模型+高剂量红景天苷组(CS+H)。首先COPD模型+安慰剂组(CS)、COPD模型+低剂量红景天苷组(CS+L)、COPD模型+中剂量红景天苷组(CS+M)、COPD模型+高剂量红景天苷组(CS+H)均采取单纯被动吸烟发建立吸烟大鼠模型,空白组采取伪暴露法建立空白组模型。空白组及COPD模型+安慰剂组(CS)给予安慰剂(生理盐水)灌胃1个月,COPD模型+低剂量红景天苷组(CS+L)、COPD模型+中剂量红景天苷组(CS+M)、COPD模型+高剂量红景天苷组(CS+H)分别予以低剂量(5mg/100g)、中剂量(25mg/100g)、高剂量(125mg/100g)红景天苷灌胃处理1个月。记录其体重变化,采用竭力游泳实验评估运动能力,采集血清、肺组织、股四头肌,对骨骼肌进行称重,测定骨骼肌样本I型纤维比例,FOXO1、P-FOXO1、Mu RF-1、Atrogin-1in-1表达水平。结果:(1)体重比较:CS组(404.6±31.7g)较C组(475.6±20.4 g)下降,差异有统计学意义(P0.05);CS+L组(419.3±20.9 g)、CS+M组(425.6±32.6g)、CS+H组(432.7±27.2g)与CS组比较,体重均有所增加,并且各组值间值差异均有统计学意义(P0.05)。(2)竭力游泳时间比较:CS组(52.4±4.5min)较C组(14.9±4.1min)缩短,差异有统计学意义(P0.05);CS+L组(22.5±2.9min)、CS+M组(23.1±2.9min)、CS+H组(31.1±4.7min)与CS组比较,竭力游泳时间均有延长,并且各组值间值差异均有统计学意义(P0.05)(3)股四头肌重量比较:CS组(2.82±0.22g)较C组(2.82±0.22g)减少,差异有统计学意义(P0.05);CS+L组(2.99±0.20g)、CS+M组(3.13±0.23g)、CS+H组(3.44±0.23g)与CS组比较,股四头肌重量增加,并且各组值间值差异均有统计学意义(P0.05)(4)I型肌纤维比例比较:CS组(16.3±4.9%)较C组(32.8±2.8%)减少,差异有统计学意义(P0.05);CS+L组(22.2±3.8%)、CS+M组(25.7±3.9%)、CS+H组(28.2±2.9%)与CS组比较,I型肌纤维比例增加,并且各组值间值差异均有统计学意义(P0.05)(5)蛋白表达比较:(1)FOXO1:C组(0.260±0.10)、CS组(0.646±0.11)、CS+L组(0.440±0.13)、CS+M组(0.380±0.12)、CS+H组(0.330±0.01);(2)P-FOXO1:C组(0.230±0.06)、CS组(0.343±0.08)、CS+L组(0.401±0.06)、CS+M组(0.314±0.07)、CS+H组(0.289±0.06);(3)Mu RF-1:C组(0.338±0.077)、CS组(0.634±0.101)、CS+L组(0.428±0.067)、CS+M组(0.530±0.065)、CS+H组(0.353±0.105);(4)Atrogin-1:C组(0.393±0.013)、CS组(0.370±0.071)、CS+L组(0.306±0.103)、CS+M组(0.286±0.005)、CS+H组(0.157±0.061)。与CS组比较,FOXO1表达有所增加,并且各组值间值差异均有统计学意义(P0.05)。CS组与C组比较FOXO1、FOXO1、P-FOXO、Mu RF-1、的表达水平升高,差异均有统计学意义(P0.05);CS+L组、CS+M组、CS+H组与CS组比较,FOXO1、FOXO1、P-FOXO、Mu RF-1、Atrogin-1的表达水平下降,且影响水平与干预剂量呈正相关,差异均有统计学意义(P0.05)。结论:1.红景天苷可能通过降低FOXO1的表达保护COPD模型大鼠的骨骼肌功能,且这种保护作用为剂量依赖型。
[Abstract]:Objective: To explore the effects of different doses of salidroside on skeletal muscle function and FOXO1 expression in COPD rats, and to provide experimental evidence for salidroside application in improving skeletal muscle dysfunction of COPD.
