粉尘螨Ⅲ类重组变应原对哮喘小鼠免疫治疗的效果

【摘要】:目的探讨粉尘螨Ⅲ类重组变应原(rDer f 3)对过敏性哮喘小鼠的免疫治疗效果。方法随机将40只BALB/c小鼠均分为4组,哮喘组、免疫治疗组、卵清蛋白(OVA)组和PBS组,分别在第0、第7和第14天哮喘组和免疫治疗组每鼠经腹腔注射100μl致敏液(含rDer f 3 10μg);卵清蛋白组每鼠经腹腔注射100μl致敏液(含OVA 10μg);PBS组则以PBS代替致敏液。第21天起,哮喘组和免疫治疗组小鼠用rDer f 3进行滴鼻激发试验,连续7 d,免疫治疗组小鼠在第25~27天滴鼻激发前30 min,用100μg rDer f 3纯化蛋白皮下注射进行特异性免疫治疗。PBS组和卵清蛋白组则分别用PBS和OVA进行滴鼻激发和腹腔注射,最后1次滴鼻激发24 h后脱臼处死小鼠。收集各组小鼠支气管肺泡灌洗液(BALF)进行白细胞和嗜酸粒细胞计数;对肺组织病理切片进行HE染色,镜下观察肺组织炎症细胞浸润情况。ELISA检测BALF和脾细胞培养上清(SSCS)中白细胞介素-5(IL-5)和γ干扰素(IFN-γ)及血清中变应原特异性IgE、IgG2a抗体水平。结果肺组织病理切片结果显示,免疫治疗组小鼠炎症反应明显减轻。小鼠BALF中白细胞总数在免疫治疗组、卵清蛋白组和哮喘组分别为(7.03±1.38)×108/ml、(22.11±3.70)×108/ml和(22.75±3.24)×108/ml,免疫治疗组低于卵清蛋白组和哮喘组(P0.01),嗜酸粒细胞的变化趋势与白细胞类似。免疫治疗组、卵清蛋白组和哮喘组BALF中IL-5的水平分别为(108.20±11.02)pg/ml、(182.04±13.94)pg/ml和(195.33±15.33)pg/ml,SSCS中IL-5的水平分别为(98.34±13.06)pg/ml、(208.26±10.63)pg/ml和(179.54±13.65)pg/ml,免疫治疗组均明显低于卵清蛋白组和哮喘组(P0.01)。而IFN-γ含量则高于卵清蛋白组和哮喘组(P0.01)。IgE水平免疫治疗组、卵清蛋白组和哮喘组分别为(9.12±3.78)IU/ml、(26.87±4.30)IU/ml和(35.25±8.84)IU/ml,与卵清蛋白组和哮喘组相比,免疫治疗组明显降低(P0.01),而IgG2a水平(38.52±6.33)μg/ml则显著升高(P0.01)。结论粉尘螨Ⅲ类变应原可逆转哮喘小鼠的变态反应性气道及肺部炎症。
[Abstract]:Objective to investigate the effect of recombinant allergen (rDer f 3 on allergic asthma mice. Methods 40 BALB/c mice were randomly divided into 4 groups: asthma group, immunotherapy group, ovalbumin (OVA) group and PBS group. On the 7th and 14th days, asthma group and immunotherapy group were injected intraperitoneally with 100 渭 l sensitization solution (including rDer f 3 10 渭 g);). In ovalbumin group, 100 渭 l sensitizing solution was injected intraperitoneally. In 10 渭 g); PBS group containing OVA, PBS was used instead of sensitizing solution. From the 21st day, mice in asthma group and immunotherapy group were given nasal drip challenge test with rDer f3 for 7 days. The mice in immunotherapy group were given nasal drip challenge 30 min, before nasal drip on day 2527. The specific immunotherapy was performed by subcutaneous injection of 100 渭 g rDer f 3 purified protein. In PBS group and ovalbumin group, PBS and OVA were used for nasal drip and intraperitoneal injection, respectively. The mice were killed after 24 hours of nasal drip. (BALF) in bronchoalveolar lavage fluid was collected to count leukocytes and eosinophils in each group. The pathological sections of lung were stained with HE. ELISA was used to detect the levels of interleukin-5 (IL-5) and interferon 纬 (IFN- 纬) in (SSCS) and serum anti-allergen specific IgE,IgG2a antibody. Results the pathological section of lung tissue showed that the inflammatory reaction in immunotherapy group was obviously alleviated. The total number of leukocytes in BALF in immunotherapy group, ovalbumin group and asthma group were (7.03 卤1.38) 脳 108 / ml, (22.11 卤3.70) 脳 108/ml and (22.75 卤3.24) 脳 108 / ml, respectively. The immunotherapy group was lower than ovalbumin group and asthma group (P0.01). The levels of IL-5 in BALF of immunotherapy group, ovalbumin group and asthma group were (108.20 卤11.02) pg/ml, (182.04 卤13.94) pg/ml and (195.33 卤15.33) pg/ml, respectively. The levels of IL-5 in SSCS (98.34 卤13.06) pg/ml, (208.26 卤10.63) pg/ml and (179.54 卤13.65) pg/ml, immunotherapy group were significantly lower than those in ovalbumin group and asthma group (P0.01). The levels of IFN- 纬 in ovalbumin group and asthma group were higher than those in ovalbumin group and asthma group (P0.01).
